摘  要:目的:探讨塞来昔布对小鼠膀胱肿瘤的抑制作用及其机制。方法:用BTT739膀胱癌细胞株和T739小鼠建立膀胱肿瘤动物模型。小鼠随机分为实验组和对照组,每组10只。造模后第2天起实验组用塞来昔布混悬液(20mg·kg-1)灌胃,对照组用0.9%氯化钠液灌胃。观察肿瘤的生长情况。4周后,处死小鼠,取肿瘤,称瘤重,计算抑瘤率。肿瘤组织行病理学检查,观察肿瘤细胞的形态学变化,用免疫组化法观察肿瘤细胞中COX-2表达情况,TUNEL法观察肿瘤细胞凋亡情况。结果:对照组平均瘤重(3.27±1.89)g,实验组平均瘤重(1.10±0.52)g,两组差别有统计学意义(P<0.05);抑瘤率为66.4%。两组肿瘤潜伏期[(7.10±2.28)d比(7.20±2.71)d]无显著差异(P>0.05)。自第2周开始两组肿瘤生长有差异,直至第4周实验结束。显微镜下见实验组有大片凝固性坏死区,而对照组仅见局灶性坏死。免疫组化示COX-2表达于肿瘤细胞的细胞浆。TUNEL法见细胞核阳性表达。两组COX-2、凋亡的表达水平均有显著差异(P<0.05),且COX-2与凋亡的表达呈负相关。结论:塞来昔布对小鼠膀胱肿瘤有抑制作用,可能通过抑制COX-2表达促进肿瘤细胞凋亡,抑制肿瘤生长。Objective:To study the inhibiting effects of celecoxib on mice bladder cancer and research the mechanism. Methods: Bladder cancer models were made with T739 mice and BTT739 cell line. All mice were randomly divided into control group and test group,with 10 in each group. From the second day after operation,mice in test group were administered orally with celeeoxib in suspension at 20 mg·kg^-1 ,while the control group were given saline. During the test,observed the growth of tumor mass and drew the growth curve. 4 weeks later,executed mice,reseeted tumors,compare their weight and were taken for routine pathology examination. The expression of COX 2 with immunohistochemistry and that of apoptosis with TUNEL were observeel. Results: The mean weight of test group was (1.10± 0.52)g, while the control group was (3.27± 1.89)g, the difference was significant in statistics (P〈0.05). The difference of latent period between test group and control group was not significant statistieally[(7.10±2.28)d vs. (7.20±2.71)d, P〉0. 05]. From the growth curve, There was difference in tumor volume between control group and test group,and it lastsed to the end of the test. Pathologically,large necrosis area could be seen in test group,while control group not. In immunohistochemistry, COX-2 was positive in cell plasma, while in TUNEL,apoptosis was positive in cell core. The expression of COX-2,apoptosis in test group was statistically different from that in control group(P〈0.05 or P〈0.01 ). And there was a negative correlation between the expression of COX-2 and apoptosis. Conclusion: Celecoxib inhibits COX-2 to induce apoptosis so as to restrain bladder cancer.
