摘  要:目的研究二烯丙基二硫(DADS)体外抑制人小细胞肺癌H446细胞增殖作用、诱导凋亡作用以及可能的分子机制。方法采用MTT、细胞计数、细胞活力检测DADS对体外培养的人小细胞肺癌H446细胞的抑制增殖作用;普通光学显微镜,荧光显微镜观察DADS对体外培养的人肺癌H446细胞的形态学改变;流式细胞仪检测DADS对H446细胞的细胞周期阻滞作用和细胞凋亡率的影响。结果MTT结果显示DADS作用于H446细胞后,能明显抑制细胞增殖且抑制率呈现浓度时间依赖性关系;细胞计数结果显示DADS作用于H446细胞后其细胞群体倍增时间延长;流式细胞仪检测结果显示DADS能诱导H446细胞凋亡。结论DADS能明显抑制H446细胞增殖并诱导其发生凋亡。Objective To investigate the effects of diallyl disulfide (DADS) on cell proliferation in human small cell lung cancer H446 cells in vitro. Methods MTT assay was used to observe inhibitory effect of DADS on proliferation of H446 cells. Cell Proliferation inhibition was measured by growth curve analysis, average doubling time, vitality detection and MTT assay. Cell morphology was observed by inversion microscope and optics microscope. Cell apoptosis was analyzed by cell morphology observed under light microscope, flow cytometry (FCM). Results MTT assay showed that DADS from 4 to 60μg/ml significantly inhibited H446 cells and exhibited a dose-dependent and time-dependent model. After exposure to DADS, H446 cell average doubling time retarded from 25.40 hours to 145.64 hours ( P 〈 0. 05). Flow cytometry analysis revealed that the cell content of G0-phase declined, however, hypodiplod peak was increased, which means cell apoptosis were induced by DADS. Conclusion DADS could significantly inhibit the proliferation of H446 cells and induce the apoptosis of H446 cell.
