白血病患者化疗前后骨髓基质细胞上清液中IL—6、TNF—α和IFN—α活性的研究

摘  要:为探讨IL—6、TNF—α、和IFN—α在化疗中的变化,测定20例完全缓解期白血病患者化疗前、后骨髓基质细胞上清液中IL—6、TNF—α、IFN—α活性,结果显示:与化疗前相比,化疗后此3者活性逐渐下降,第14 d时IL—6下降至最低;第12 d时TNF—α、IFN—α下降至最低(P<0.01)。随着化疗时间的延长,IL—6逐渐恢复正常,TNF—α、INF—α的活性也逐渐升高,且三者的变化与白细胞值的变化呈正相关。提示:基质细胞上清液是IL—6、TNF—α和IFN—α活性反应了基质细胞损伤的程度,可以作为白血病患者化疗后骨髓造血功能恢复和免疫重建的指标。IL-6, TNF-α and IFN-α activities were detected in stromal cell supernatant before and after chemotherapy in 20 leukemic patients who acquired complete remission. The results showed that the activities of IL-6, TNF-α and IFN-α gradually reduced after chemotherapy compared with pre—chemotberapy. IL--6 activity reduced to the lowest level at the 14th day after chemotherapy while TNF-α and IFN-α activities did at the 12th day (P<0.01). With the chemotherapy time prolonged, IL—6 activity gradually elevated to normal level, so did the TNF-α and IFN-α activities. All the changes of the activities of the three factors corelated positively with the number of the white blood cells. These results suggested that the activities of IL-6, TNF-α and IFN-α in stromal cell supernatant reflect tbe degree of stromall cell injury, which can be used as an index to indicate bone marrow hemotopoietic restoration and immune rebuilding after chemotherapy of leukemic patients.
