摘 要:目的初步探讨白细胞介素-1受体拮抗剂(IL-1ra)基因(IL-1RN)多态性与安徽地区胃癌易感性的关系,并比较不同种族和地区之间IL-1RN*2等位基因频率的分布。方法采用聚合酶链式反应(PCR)技术检测安徽地区91例胃腺癌患者及55例健康对照者IL-1RN基因多态性。结果IL-1RN*2*2基因型频率及IL-1RN*2等位基因频率在胃癌组与对照组的比较,差异无显著性(P>0.05);在不同种族的比较中,IL-1RN*2等位基因频率在本研究人群显著低于欧洲高加索人群,不论在胃癌组还是对照组(15.4%vs57.4%,14.5%vs44.8%)(P<0.01);在胃癌组略高于日本人群(15.4%vs7.7%)(P<0.05)。在中国不同地区的比较中,IL-1RN*2等位基因频率在本研究人群(安徽地区)显著低于北部地区人群,不论在胃癌组还是对照组(15.4%vs76.0%,14.5%vs46.0%)(P<0.01);在对照组中略高与西部地区人群(14.5%vs3.6%)(P<0.05);与东部及南部地区比较,差异无显著性。结论IL-1RN*2*2基因型及IL-1RN*2等位基因在胃癌高发区安徽地区与胃癌的易感性无关。Objective To investigate the relationship between Interleukin-1RN gene polymorphism and the risk of gastric cancer in Anhui province,and find the difference of frequency of IL-1RN2 allele between population with different races and different areas.Methods Fifty-five health controls and ninety-one patients with gastric cancer in Anhui province were genotyped by polymerase chain reaction(PCR)for 86 bp variable number of tandem repeat(VNTR)polymorphism in IL-lRN gene intron 2(for five different alleles).Result No significant difference in the frequency of IL-1RN22 and IL-1RN2 was found between gastric cancer patients and controls in our population(P〉0.05),but a significant difference in the frequency of IL-1RN2 was found between our population and Caucasian population in both gastric cancer patients and controls(15.4% vs 57.4%,14.5% vs 44.8%),(P〈0.01);a little difference was found between our population and Japanese population in gastric cancer patients(15.4% vs 7.7%),(P〈0.05).Compared with other areas in China,the frequency of IL-1RN2 in our population(Anhui province,central area of China)was significant different from that in north area in both gastric cancer patients and controls(15.4% vs 76.0%,14.5% vs 46.0%),(P〈0.01).There was a little different from that in west area in control population(14.5% vs 3.6%)(P〈0.05);but no significant difference was found between our population and east and south population.Conclusion IL-1RN gene polymorphism may be not associated with the risk of gastric cancer in this high-risk area.
