摘  要:目的:观察姜黄素(curcumin)对人食管癌Ec-9706细胞凋亡作用及凋亡抑制蛋白livin,促凋亡蛋白Smac和caspase-3蛋白表达的影响,探索其分子机制.方法:应用40~160μmol/L姜黄素处理Ec-9706细胞12~48h后,采用四甲基偶氮唑蓝法(MTT)检测细胞增殖抑制率;流式细胞术(FCM)检测Ec-9706细胞凋亡率:免疫蛋白印迹技术(WesternBlot)检测livin,Smac及caspase-3蛋白的表达.结果:姜黄素作用后Ec-9706细胞生长明显减慢,抑制率9.3%~73.2%(P〈0.01),凋亡率为6.0%~45.2%(P〈0.01),并呈良好的时间-剂量效应关系.药物作用后,Smac,caspase-3蛋白表达升高,livin蛋白表达下调(P〈0.01).结论:姜黄素能明显抑制Ec-9706细胞的增殖,诱导其凋亡.其机制可能与抑制livin蛋白表达,同时促进Smac,caspase-3表达有关.AIM: To explore the effect of curcumin on the induetion of apoptosis in the esophageal epithelia Ee-9706 cells and livin, Smae, easpase-3 protein expressions, and study the molecular mechanism. METHODS: After Ee-9706 cells were treated with 40 - 160 ~mol/L eureumin for 12 -48 h, Methyl thiazolyl tetrazo- lium (MTY) method was used to deteet the inhibition rates. Flow eytometry (FCM) was employed to detect Ee-9706 eeU apoptosis rates. The expressions of livin, Smae, easpase-3 proteins were analyzed by Western Blot. RESULTS : After eureumin treatment, the growth of Ec-9706 cells was inhibited significantly in a time- and dose-dependent manner, and the inhibition rates were 9. 3% - 73.2% ( P 〈 0. 01 ), apoptosis rates were 6. 0% - 45.2% ( P 〈 0. 01 ). The expressions of Smac ,caspase-3 proteins were up -regulated, and livin was down-regulated (P 〈 0. 01 ). CONCLUSION: Curcumin can inhibit the proliferation and induce the apoptosis of Ec-9706 cells. Its possible molecular mechanism might be related to the modulation of livin, Smac and caspase-3 protein levels.
