摘  要:目的:研究脑星形细胞肿瘤中Survivin基因mRNA表达的临床、病理和预后的关系。方法:应用RT-PCR方法检测26例脑星形细胞肿瘤组织中Survivin基因mRNA的表达,并通过电泳凝胶图像系统检测Survivin的相对表达量。结果:Survivin只表达于脑星形细胞肿瘤中的肿瘤细胞,对正常脑组织中胶质细胞无表达。在脑星形细胞肿瘤中RT-PCR的检出率为65%(17/26)。Survivin基因mRNA的相对表达量与脑星形细胞肿瘤的病理分级密切相关,在高级别肿瘤中Survivin基因mRNA相对表达量和表达强度均高于低级别肿瘤(P<0.05)。结论:随着星形细胞肿瘤分化程度的降低,Survivin相对表达量显著增强;其高表达预示着预后不良,可成为有价值的评估该肿瘤预后指标。Objective:To investigate the relationships between the expression of Survivin gene (at mRNA levels)and the clinico- pathologic parameters in astroeytomas ,and evaluate its significance in predicting the prognosis.Methods:The expression of Survivin mRNA in 26 fresh astrocytomas was examined by RT-PCR. The relative expression of Survivin by electrophoresis gel image system was calculated. Results:The expression of Survivin was only found in tumor eells in astroeytomas and there was no expression in normal brain tissues. The positive rate of RT-PCR was 65%(17/26).The relative expression of Survivin by RT-PCR had a high correlation with pathological grades of astrocytomas. The relative expression and expression intensity of Survivin gene in high-grade tumors were higher than those in low-grade tumors (P〈0.05).Conclusion :The relative expression and expression intensity of Survivin gene reinforced sig- nificantly when the differentiation degree decreased. Survivin might play an important role in astroeytomas conversion process from a low degree to a higher degree. Survivin can become a valuable evaluation marker of prognosis.
