摘  要:目的:本实验旨在研究炎症因子高迁移率族蛋白B1(High mobility group box protein1,HMGB1)在肝纤维化中的表达水平及其与肝纤维化发生发展之间的关系。方法:采用复合因素制备肝纤维化模型。选用雄性Wistar大鼠30只,随机分为两组:对照组和模型组,分别在饲养第0、2、4、6、8周末取肝组织做免疫组织化学染色并采用Image-ProPlus5.0图像分析系统对HMGB1的阳性表达进行图像分析,计算累计光密度值(IOD),以观察肝组织HMGB1的表达水平。采血测定外周血清中的透明质酸(HA)、丙氨酸氨基转移酶(ALT)的动态表达水平变化。结果:外周血清ALT及肝组织中HMGB1的IOD值在第2周末达到高峰,随后下降,到第8周末仍高于正常组(P<0.05);外周血清HA在第4周末达到高峰随后下降,到第8周末仍高于正常组(P<0.05)。结论:HMGB1作为炎症因子与肝纤维化的发生发展密切相关。Objective:To investigate the expression of High Mobility Group Box 1 in the rat liver fibrosis induced by CCl4 and the relationship between the expression of HMGB 1 and rat liver fibrogenesis. Methods : Thirty male Wistar rats were randomized into two groups (control group, model group), in which all rats were given the subcutaneous injection of 40% CCL4(every 3 days for 2.4.6.8weeks ) except for rats of control group.After 2. 4.6.8 weeks liver tissue and serum samples of all rats were examined. Serum hyaluronic acid (HA) were detected by Radioimmunoassay and ALT were detected by LAISHI technique. The expression of HMGB 1 in liver tissue were determined by immunohistochemical techniques and were analyzed semiquantitatively by image - analyzing system. Results:Compared with control group, the serum ALT of model groups remarkbably increased especially at the 2th weekend and the serum HA of model groups reached its peak at the 4th weekend. The maximum IOD of HMGB 1 of model groups were also detected at the 2th weekend. The expression of ALT, HA and HMGB 1 of model groups almost show a similar change tendency. Conclusion: It is suggested that HMGB 1 is related with liver fibrosis of rats closely and may be a fibrogenesis factor of liver.
