摘  要:目的:观察表没食子儿茶素没食子酸酯(EGCG)抑制人卵巢癌(CoC1)细胞系,细胞生长的生物学效应。方法:通过MTT比色法检测EGCG对CoC1细胞生长的影响;应用细胞计数法研究不同浓度EGCG对CoC1细胞生长曲线的影响;采用平皿克隆形成实验法测定不同剂量EGCG对人卵巢癌(CoC1)细胞锚定依赖性生长能力的影响。结果:EGCG显著抑制人卵巢癌CoC1细胞的生长,呈浓度依赖性;CoC1细胞经EGCG作用后,其细胞群体倍增时间明显延长从正常生长的38.02h,直至97.56h,细胞增殖周期延长,从而细胞增殖速度减慢;平皿克隆形成实验法显示:EGCG可抑制人卵巢癌细胞CoC1生长,且呈剂量依赖性。结论:EGCG具有抑制人卵巢癌CoC1细胞生长的作用,其机制可能与细胞增殖周期延长相关。Objective:To investigate the effect of epigallocatechin - 3 - gallate on Inhibition of Proliferation in Ovarian cancer cell line CoC 1. Methods : The growth inhibition of CoC 1 cells was measured by MTF assay. Cell couting method was used to test the influence of growth curve in CoC 1 ceils treated with different concentrations of EGCG. Cell proliferation of CoC 1 cells was evaluated by clone formation test. Resuhs:EGCG significantly inhibited the growth of CoC 1 cells, in a concentration dependent manner. Cell counting showed that average doubling time retarded from33.06 h in normal culture CoC 1 cells to 38.02 h in 20μg/mL EGCG, even to 97.56 h in 100μg/mL EGCG experimented CoC1 cells. Colone formation test on CoC1 cells indicated that EGCG could inhibit the proliferation of CoC 1 cells in a concentration - dependent manner. Conclusion : EGCG posseses antitumot effect in vitro, its mechanism might be associated with doubling time retarded.
