摘 要:目的:运用骨密度、骨生物力学检测和Micro CT分析及三维结构重建的方法,观察健骨汤对老龄去势大鼠松质骨的影响。方法:30只12月龄雄性wistar大鼠随机分为ORX组、Sham组、ORX加健骨汤(ORX+JG)组,每组10只。前两组生理盐水灌胃每日,ORX+JG组用健骨汤提取液(5ml/kg/d))灌胃,12周后取材.运用双能X线骨矿含量测定仪(DXA)检测治疗后大鼠全身总骨密度;取L3椎体、右股骨行MicroCT分析及三维结构重建,取L4椎体、左股骨行骨生物力学测试。结果:与去势组比较,ORX+JG组BMD、骨小梁体积(BV/TV)、骨小梁厚度(Tb.Th)、骨小梁数目(Tb.N)值升高,骨生物力学检测发现骨最大载荷(M-load)、吸收能量(Enery)、最大应力(M-stress)、弹性模量(E-lastic)值升高,骨小梁间隙(Tb.Sp)、结构模型指数(SMI)值降低。结论:健骨汤能明显改善去势大鼠骨微结构,增加骨强度和骨密度,防止骨量丢失。Objective:To investigate the influence of Jiangu Decoction (JGD) on cancellous bone in senile orchiectomized (ORX) rats via bone mineral density (BMD) test, bone biomechanical analysis, micro--CT scanning and three--dimensional structure reconstruction. Methods: Thirty 12--month--old male Wister rats were randomly divided into ORX group, sham--operated group (Sham group) and ORX+JGD group, 10 rats in each group. Rats in Sham group and ORX group received intragastric administration of 0, 9% NaCl daily while rats in ORX+JD group received JGD extract (Sml/kg/d) for 12 weeks. Dual energy X--ray absorptiometry was used to measure total body BMD, micro--CT scanning and three--dimensional structure reconstruction were used to analyze right femurs and L-3 vertebra, and bone biomechanical test was carried on left femurs and L-4 vertebra. Results: Compared with ORX group, the following indexes in ORX+JGD all significantly increased: BMD, the volume, thickness and number of trabeeular bone, maximum load, energy absorption, maximum stress and elastic modulous. Meanwhile, the space of trabecular bone and structure model index all decreased. Conclusion:Jiangu Decoction could prevent the bone loss, increase micro-architecture and biomeehanical properties of the trabecular bone in ORX rats.
