摘 要:目的探讨不同延迟时间点给予亚低温治疗对永久性大脑中动脉阻塞大鼠的神经保护效应。方法雄性SD大鼠30只,线栓法制备永久性大脑中动脉阻塞模型,随机分为亚低温治疗2h组(HT2h组)、亚低温治疗6h组(HT6h组)和缺血对照组(NT组),每组10只。应用激光多普勒监测局部大脑中动脉供血区脑血流量。持续监测核心温度。取脑制备冷冻切片,进行焦油紫染色,计算脑梗死体积、脑水肿程度,TUNEL染色确定神经元凋亡情况。结果与NT组比较,HT2h组和HT6h组神经功能缺损评分、脑梗死体积/全脑体积、脑水肿程度以及缺血半暗带区域内大脑皮质区、纹状体区和丘脑区TUNEL阳性细胞计数均显著降低。与HT6h组比较,HT2h组脑梗死体积全脑体积、脑水肿程度以及缺血半暗带区域内大脑皮质区和纹状体区TUNEL阳性细胞计数均显著降低。结论延迟亚低温治疗通过抑制大脑中动脉阻塞后神经细胞凋亡,从而达到神经保护作用。Objective To explore the neruoprotective effect of the delayed mild hypothermia on the rats with permanent cerebral ischemia. Methods Thirty male SD rats were used to create permanent middle cerebral artery occlusion models by intraluminal suture of the artery. The animals were randomly assigned to 3 experimental groups. Mild hypothermia[(33.0±0. 5)℃] was achieved after 2 h(HT 2 h group) and 6 h(HT 6 h group),and was maintained for 22 h and 18 h respectively,control group was raised at room temperature. Local cerebral blood flow in the supply territory of the middle cerebral artery of the crerbral cortex was monitored by laser-Doppler flowmetry. The rectal temperature was continually monitored. The brains were removed and cut into sections for cresyl violet(CV) staining and TUNEL staining. The infarct volume and brain edema were measured by CV staining. Results The Longa's score,infarct volume, brain edema,and quantity of apoptotic neurons in penumbra of mild hypothermic groups were significantly decreased compared with control group. There was significant decrease in the infarct volume, brain edema and quantity of apoptotic neurons in penumbra of cortex and striatum in HT 2 h group compared with HT 6 h group. Conclusions The delayed mild hypothermia produced a marked effect of neuroprotection by decreasing apoptosis of neurons in the penumbra region.
