摘  要:背景:人端粒保护蛋白1(hPOT1)的主要作用为保护端粒和调节端粒长度。目的:探讨hPOT1在人胃癌细胞中对端粒长度的调节作用及其可能机制。方法:以RNA干扰(RNAi)技术抑制人胃癌细胞株BGC-823中hPOT1的表达,以实时荧光定量聚合酶链反应(PCR)检测细胞端粒长度,以半定量逆转录聚合酶链反应(RT-PCR)检测人端粒酶逆转录酶(hTERT)mRNA表达。结果:以RNAi技术抑制hPOT1表达后,BGC-823细胞端粒长度明显缩短,反映端粒相对长度的T/S值显著小于亲本BGC-823细胞组和阴性对照组(1.383±0.091对2.758±0.647和3.043±0.548,P<0.05),亲本细胞组与阴性对照组之间则无明显差异。hPOT1 RNAi组细胞hTERT mRNA表达亦明显下调。结论:在人胃癌细胞中,hPOT1能正性调节端粒长度;在hPOT1下调引起端粒缩短的环节中,hTERT表达下降可能起一定作用。Protection of telomere and regulation of telomere length are the two main functions of human protection of telomeres 1 (hPOT1) protein. Aims: To investigate the effect of hPOT1 on regulation of telomere length in human gastric cancer ceils and its potential mechanism. Methods: Expression of hPOT1 in human gastric cancer cell line BGC-823 was inhibited by RNA interference (RNAi). Telomere length of BGC-823 cells was measured by real-time fluorescent quantitative polymerase chain reaction (PCR), and expression of human telomerase reverse transcriptase (hTERT) mRNA was determined by semi-quantitative reverse transcriptase polymerase chain reaction (RT-PCR). Results: Telomere length of BGC-823 cells in hPOT1 RNAi group was significantly shorter than that of parent BGC-823 cells and negative controls (P〈0.05), whereas no significant difference was observed between parent cells and negative controls. T/S ratios (reflecting relative telomere length) in hPOT1 RNAi group, parent BGC-823 cells group and negative control group were 1.383±0.091, 2.758±0.647 and 3.043±0.548, respectively. Expression of hTERT mRNA was also down-regulated significantly in hPOT1 RNAi group. Conclusions: hPOT1 regulates the telomere length positively in human gastric cancer cells. Decrease of hTERT expression may be involved in the telomere shortening induced by hPOT1 down-regulation.
