OPN和uPA mRNA在肝细胞癌组织中的表达及临床意义

摘　　要：目的:探讨骨桥蛋白(OPN)mRNA及尿激酶型纤溶酶原激活物(uPA)mRNA在肝细胞癌(HCC)组织中的表达及与临床病理特征的关系。方法:应用RT-PCR方法检测60例HCC癌组织、20例正常对照肝组织中OPN和uPA mRNA的相对表达量。结果:HCC癌组织中OPN和uPA mRNA的阳性表达率均分别高于对照正常肝组织(P均<0.05);癌组织中OPN mRNA的表达量在有门静脉癌栓形成组及在有早期复发转移组均高于无门静脉癌栓形成组和无早期复发转移组(分别P<0.05和P<0.01),uPA mRNA的表达水平在有包膜浸润和有门静脉癌栓形成组中均高于无包膜浸润和无门静脉癌栓形成组(分别P<0.05和P<0.01);OPN mRNA的高表达和uPA mRNA的高表达呈正相关(P<0.05)。结论:HCC癌组织中OPN的表达水平可以作为判断肝癌细胞转移能力和预示肝癌早期复发的重要指标。阻断OPN的表达可能可以降低uPA的分泌,从而减少肿瘤的浸润和转移。Objective：To investigate the expression of OPN mRNA,uPA mRNA and their correlations with the clinical pathological characteristics in hepatocellular carcinoma （HCC）. Methods：60 cancer tissues and 20 normal tissues was semi-quantitively detected by RT-PCR. Results ：The positive expression rates of OPN and uPA mRNA in HCC tissues are both higher than that in control tissues （bothP 〈 0.05）;In HCC tissues,the expression of OPN mRNA were both higher in the cases with early recurrence or metastasis and with portal vein tumor embolus than that in the cases without（respectively P〈0.05 and P〈0. 01） ,the expression of uPA mRNA were both higher in the cases with peplos infiltration and with portal vein tumor embolus than that in the cases without （respectively P〈0. 05 and P〈0. 01）; Higher expression of OPN mRNA was positively correlated with higher expression of uPA mRNA（P〈0.05）. Conclusions：OPN is an important marker to judge the potential ability of metastasis and to indict early recurrence or metastasis of HCC. Perhaps to block the expression of OPN may reduce the secretion of uPA,thus may decrease the tumor＇s infiltration and metastasis.
