摘  要:目的探讨白藜芦醇对MDA-MB-231细胞的凋亡诱导效应和对细胞内活性氧水平(reactive oxygen species,ROS)及Caspase-3的影响。方法用不同浓度的白藜芦醇诱导培养MDA-MB-231细胞24h后,采用流式细胞术(FCM)和DNA琼脂糖凝胶电泳检测细胞凋亡,FCM测定细胞内活性氧(ROS)水平,免疫组织化学法检测Caspase-3蛋白的变化。结果白藜芦醇能诱导MDA-MB-231细胞凋亡,DNA电泳可见梯状条带出现,并呈现典型的凋亡改变,60μmol/L白藜芦醇作用MDA-MB-231细胞4、12和24h后细胞ROS生成量分别为:50.0±5.7、115.0±10.6、79.2±6.9;Caspase-3蛋白活性逐渐增加。结论白藜芦醇诱导MDA-MB-231细胞凋亡与细胞ROS水平升高和Caspase-3活化有关,可能是白藜芦醇诱导MDA-MB-231细胞凋亡机制之一。Objective To investigate the apoptosis of MDA-MB-231 cells induced by resveratrol and the influence resveratrol on the intracellular reactive oxygen species(ROS)level and Caspase-3.Methods MDA-MB-231 cells were treated with resveratrol in different concentrations for 24 h.Flow cytometry(FCM)and DNA agarose gel electrophoresis were used to detect the apoptosis of MDA-MB-231 cells.Intracellular ROS level was measured with FCM,and the expression of Caspase-3 was determined by immunohistochemistry.Results Resveratrol could induce the apoptosis of MDA-MB-231 cells.Agarose gel electrophoresis showed characteristic DNA ladder fragmentation,as well as the typical change of apoptosis.The level of ROS in MDA-MB-231 cells were(50.0±5.7),(115.0±10.6)and(79.2±6.9),respectively,after treatment by 60 μmol/L resveratrol for 4h,12h and 24h.Caspase-3 was also activated by Resveratrol.Conclusion Resveratrol induced apoptosis is related to the increased ROS and activated Caspase-3 in MDA-MB-231 cells.It is may be one of the mechanisms of apoptosis induced by Resveratrol in MDA-MB-231 cells.
