摘  要:目的探讨COX-2、VEGF在喉鳞状细胞癌发生发展及侵袭转移中的作用。方法应用免疫组化Elivision法检测30例喉鳞状细胞癌、10例声带非典型增生中COX-2、VEGF的表达,并在显微镜下观察微血管密度(MVD)。结果VEGF在30例喉鳞状细胞癌和10例声带非典型增生中的阳性表达率分别为86.7%,20%;COX-2在两组中的阳性表达率分别为76.7%、10%;喉鳞状细胞癌及声带非典型增生微血管密度分别为65.85±13.06、15.37±3.47。COX-2、VEGF阳性表达和微血管密度两组间差异有统计学意义。结论COX-2、VEGF在喉鳞状细胞癌中的高表达,提示有新生血管化的发生,可能与喉鳞状细胞癌的发生、发展及侵袭转移有关。选择性COX-2、VEGF抑制剂可望成为基因治疗喉鳞状细胞癌新的靶点。Objective To investigate the role of eyclooxygenases-2 ( COX-2 ) and vascular endothelial growth factor (VEGF) in the occurrence, development, and matastasis of laryngeal squamous cell carcinoma. Methods The immunohistochemical Elivision method was used for detecting the expression of COX- 2 and VEGF in 3 0 specimens of laryngeal squamous cell carcinoma and 1 0 specimens of vocal cord atypical hyperplasia , their mierovaseular density ( MVD ) was measured as well. Results The positive rates of COX - 2 were 7 6.7 % and 10 % in laryngeal squamous cell carcinoma group and vocal cord atypical hyperplasia group respectively, while those of VEGF were 86. 7% and 20 %. The differences of positive rates of COX-2, VEGF, and MVD between the two groups were statistically significant. Conclusion The high expressions of COX-2 and VEGF indicate abnormal angiogenesis in laryngeal squamous cell carcinoma, which suggests that COX-2 and VEGF play an important role in the incidence, development, and the metastasis of laryngeal squamous cell carcinoma and that specific COX-2 and VEGF inhibitors may be new targets for the gene-therapy.
