摘  要:目的探讨小鼠骨髓单个核细胞培养出的耐受性树突状细胞(tDCs)输注对免疫介导性再生障碍性贫血模型小鼠骨髓造血功能的影响。方法采用昆明小鼠30只,参照姚军等方法建立免疫介导性再生障碍性贫血模型,实验分模型组、tDCs组、照射组、正常组。tDCs组分别于制模后0d、3d、5d从小鼠尾静脉输注耐受性树突状细胞1×106/只,观察各组小鼠外周血象变化、濒死时(或造模28d时)骨髓有核细胞数量和骨髓病理学特征。结果模型组外周血WBC显著低于tDCs组和照射组,分别为(0.7±0.2)×109/L,(4.27±0.64)×109/L,(5.89±0.34)×109/L。tDCs组小鼠单侧股骨有核细胞数量显著高于模型组,而与照射组结果一致。结论tDCs输注可减轻再生障碍性贫血模型小鼠骨髓造血衰竭程度。Objective To explore the effects of infusion with tolerogenic dendritic cells on bone marrow hematopoietic function and survival in mice with aplastic anemia (AA).Methods Immuno-mediated aplastic anemia mice model was established according to Yao's method. Thirty kunming female mice were divided into AA model group,tDCs group,radiation group and normal group.In tDCs group,1 ×10^6 tDCs were infused by intravenous injection at 0 days,3 days,5 days after establishment of AA model.The changes of blood count,the number of nuclear cells and pathological features of bone marrow were observed in all groups.Results The number of white blood cells in peripheral blood of AA model group was (0.7±0.2)×10^9/L,which was significantly lower than those in tDCs group and radiation group, respectively. In tDCs group,the number of nucleated cells in single thigh bone and pathological slice were significantly higher than those in AA model group,respectively.There were no significant differences between tDCs group and radiation group in bone marrow hematopoiesis.Conclusion tDCs infusion reduced the degree of bone marrow failure of immuno-mediated AA mice model.
