摘 要:目的豚鼠镜片诱导型近视眼取后极部巩膜测定转化生长因子β2(transforming growth factor β2,TGF-β2)、结缔组织生长因子(connective tissue growth factor,CTGF)的表达,对近视眼的发病机制进行探讨。方法2周龄豚鼠10只用镜片诱导方法随机选取1眼制备近视动物模型,实验前后检测双眼屈光度、眼轴长度。造模45 d后,观察后极部巩膜的光镜电镜变化,并对TGF-β2、CTGF的表达进行定性及半定量分析。结果45 d镜片诱导后,实验眼形成明显近视,诱导出(-8.48±0.80)D的相对近视,眼轴相对延长(1.92±0.19)mm,实验前后差异有显著统计学意义(P<0.001)。光镜电镜观察实验眼发生退行性改变。TGF-β2表达实验眼多于对照眼,2组比较差异有显著统计学意义(P=0.008)。CTGF的表达2组比较差异无统计学意义(P=0.678)。TGF-β2与CTGF经统计学分析无相关性(r=0.130,P=0.720)。结论实验性近视眼发生的退行性改变是其发生发展的组织病理学基础;TGF-β2与实验性近视的形成有关;CTGF在实验性近视的形成中无明显作用,TGF-β2与CTGF在近视的形成中无明显关系。Objective To observe the expression of TGF-β2 and connective tissue growth factor (CTGF) in the posterior scleral bands of guinea pig lens-induced myopia, to discuss the pathologic mechanisms of experimental myopia. Methods Ten two-week-old guinea pigs were monocularly fitted with lenses randomly. Before and after the experiment, diopter and the length of axis oculi were measured. After 45 days, they were taken histological experiments respectively to observe the expression of TGF-β2, CTGF, the change of pathology and ultra-structure of the posterior scleral bands, which were also qualitative and semi-quantitative analyzed. Results The experimental eyes became myopia and axis oculi elongated after insertion of the lens at 45 days. Eye became myopic on average of ( - 8.48±0.80) D, axis oculi elongated on average of ( 1.92±0. 19)mm. There was obvious difference between them(P 〈0.001 ). Experimental myopia had a series of degenerative changes by observing the pathological morphology and ultra-structure. The expression of TGF-β2 was higher in experimental eye than that in control eye( P =0.008 ). Expression of CTGF was almost the same between them. No significant difference was occurred between them (P = 0.678 ). There was no correlation between TGF-β2 and CTGF for the expression of the posterior sclera with statistical analysis( r = 0. 130,P = 0. 720). Conclusions Retrogressive changes are histopathologic basis for the happening of experimental myopia. TGF-β2 plays a role in the formation of experimental myopia,but CTGF has no significant effect in experimental myopia. There was no obvious relationship between the two cytokines.
