摘  要:目的观察新生猪牙胚的发育情况并培养猪牙乳头细胞,对传代细胞的生物学特性进行研究。方法选择3日龄新生猪,分离牙胚,通过组织学、免疫组化等研究牙胚的发育情况。分离牙乳头组织,酶消化法培养牙乳头细胞并鉴定;观察细胞生长特性,通过牙本质涎蛋白、Ⅰ型胶原、Ⅲ型胶原免疫组化染色比较体内外细胞的生物学性状。结果新生猪磨牙尚未萌出,其中第二磨牙为典型的钟状期牙胚。分离培养的牙乳头细胞在体外生长良好,经鉴定细胞来源于间充质组织,免疫组化表明,牙本质涎蛋白和Ⅰ型胶原表达阳性,Ⅲ型胶原表达阴性。结论新生猪可以提供处于钟状期的牙胚,通过酶消化法可成功培养猪牙乳头细胞,第3代细胞的生物学特性与体内细胞有一定相似性,可用于进一步深入研究。Objective To investigate the development of teeth ,to culture the porcine dental papilla cell and study its eytobiological eharacheristics. Methods 3 days postnatal pigs were chosed, and teeth buds were extracted and examnined by histological and immunohistochemical methods. Dental papillae were collected and dissociated by enzyme digestion, then pDPCs were cultured in vitro in the medium of DMEM/F12 contain 10% FBS. Then immunochemistry method was performed to detect the expression of DSP, Col Ⅰ , and Col Ⅲ. Results There're no molars erupted in the mouth of 3 days old pigs. The 2nd molar was typically at bell stage, pDPCs were sueessfully cultured in vitro, and origined from menehenchymal tissues identified by Vim and CK. The immunochemistry results showed that DSP and Col Ⅰ were positively expressed and Col Ⅲ was negative. Conclusion Teeth buds at bell stage can be harvested from 3 days old pigs, and pDPCs were sucessfully cultured. The pDPCs of 3^rd generation were similar to the cells in dental papilla at a certain extent, and could be used in further study.
