摘 要:目的观察张应力与压应力两种不同应力形式对成骨样细胞增殖的影响。方法本研究应用Forcel四点弯曲体外细胞力学加载仪(ZL01256849.X)对人成骨样细胞株MG63加载,使用倒置相差显微镜观察不同力学加载强度对细胞贴壁生长的影响,应用流式细胞计量术检测生理水平周期性单轴张、压应力刺激下成骨样细胞MG63细胞周期及增殖指数的变化,揭示不同应力形式对成骨样细胞增殖的影响。结果细胞贴壁生长与应力水平相关,生理水平应力刺激下细胞贴壁及形态良好,超生理水平应力刺激不利于细胞贴壁生长。生理水平应力2000μstrain,0.5Hz加载1小时张力组细胞增殖指数上调,压力组变化不显著。结论力值大小与细胞贴壁生长密切相关;不同形式的应力对成骨样细胞增殖的影响不同,应力加载早期,张应力促进成骨样细胞增殖,压应力影响不明显。Objective To investigate osteoblast proliferation after different mechanical stimulation. Methods Osteoblasts ( MG63 ) were subjected to mechanical tension/compress strain with four - point bending system (ZL01256849. X) at 0.5Hz frequency. Strain ranged from 2000μstrain to 8000μstrain. We observed osteoblasts appearance before and after the stimulation. The ceils were subjected to physiologic magnitude tension/compress at 2000μstrain for 1h, 3h and 6h, respectively. The flow cytometry method was used to examine the osteoblast proliferation after loading. Results The cell adhesion and growth were very good after physiological stress, while the cells fell off and died after the magnitude of loading over the physiological level. After 2000μstrain tension loading for 1h, the SPF and PI of the osteoblasts promoted significantly, but compress groups had no obvious change. Conclusion Mechanical tension at physiologic magnitude could promote the proliferation of osteoblasts.
