DIM对家兔PTA术后血管内膜增生及相关生长因子的影响

摘  要:目的观察3,3′-二吲哚甲烷(3,3′-Diindolylmethane,DIM)对家兔颈动脉球囊损伤后内膜增生的影响及其作用机制。方法建立动脉球囊损伤模型。4wk后全部将兔处死,行HE染色观察动脉形态学的变化,同时进行血管内皮生长因子(vascular endothelial growth factor,VEGF)碱性成纤维细胞生长因子(basic fibroblast growth factor,bFGF)的免疫组化检测。结果损伤动脉内膜厚度(intimal thickness,IT)、中膜厚度(medial thickness,MT)及IT/MT比明显增加。与模型组比,小剂量DIM组的IT及IT/MT、VEGF与bFGF表达阳性率均有所下降,但是差异无显著性(P<0.05),而大剂量DIM组结果明显改善,差异具有显著性(P<0.01)。结论DIM可抑制血管成形术(percutaneous transluminal an-gioplasty,PTA)后再狭窄的形成及VEGF与bFGF的表达,因而可防治VSMC的增殖和新生内膜的形成。Aim To observe the effects and mechanism of DIM on neointimal proliferation of rabbit carotid artery after balloon injury. Methods Artery balloon injury model was made. The rabbits were all killed after 4 weeks. The artery morphology alteration were observed by HE dyeing, and meanwhile the vascular endothelial growth factor (VEGF) and basic fibroblast growth factor (bFGF)were detected by immunohistochemical assay. Results The injury artery intimal thickness, medial thickness and the ratio of IT/MT increased evidently. Compared with model group, the in-timal thickness decreased in low-dose DIM group, so did the IT/MT and the expression of VEGF and bFGF, but no significant difference was found ( P 〉 0. 05 ), while the results of the high-dose DIM group improved evidently with significant difference( P 〈0. 01 ). Conclusion DIM can refrain the restenosis after angioplasty and decrease the expression of VEGF and bFGF, so it can prevent and treat the proliferation of VSMC and the formation of new intima.
