摘 要:目的建立大脑皮层细胞条件培养液促进骨髓间充质干细胞向神经样细胞分化的方法。方法原代培养并鉴定骨髓间充质干细胞;用条件培养基、无血清培养基、DMEM分别诱导骨髓间充质干细胞6h和24h;用免疫荧光染色的方法鉴定由骨髓间充质干细胞分化而来的神经样细胞。结果骨髓间充质干细胞表达CD106、CD90、CD29和CD44标记物,不表达CD71、CD45和CD34,回收得到的条件培养基可以诱导骨髓间充质干细胞分化为神经样细胞,这些神经样细胞的免疫荧光染色呈β微管蛋白Ⅲ、胶质原纤维酸性蛋白、巢蛋白阳性,且阳性比例明显高于实验对照组,差异有高度统计学意义(P<0.001)。结论大脑皮层细胞条件培养液能促进骨髓间充质干细胞向神经样细胞的分化。Objective To establish the method of that conditioned medium from cerebral cortex cultures promote neurogenic differentiation of bone marrow mesenchymal stem cells(BMSCs). Methods Primary culture and identification of BMSCs: BMSCs were induced by conditioned medium, serum-free medium and DMEM for 6 h or 24 h; The differentiated neuron-like cells derived from BMSCs were characterized by specific immunofluorescence staining. Results BMSCs at passage five were positive for CD106,CD90,CD29 and CD44, negative for CD71,CD45 and CD34. The conditioned medium retrieved from the mixed cultures of newborn rat cerebral cortex could induce the BMSCs to differentiate into neuron-like cells. These neuron-like cells expressed β-Ⅲ-tubulin,GFAP and nestin, characteristics of neurons,astrocytes and neural progenitors respectively and the percentage of the positive cells was obviously higher than that of the controls (P〈0.001). Conclusion Conditioned medium from cerebral cortex cultures can promote neurogenic differentiation of BMSCs.
