动物源性ω-3 PUFAs对耐多柔比星人卵巢癌细胞系A2780逆转耐药性研究

摘  要:目的:通过人卵巢癌细胞及多柔比星耐药株的体外细胞毒性实验,探讨两种动物源性ω-3多不饱和脂肪酸对耐多柔比星人卵巢癌细胞系A2780逆转耐药性的作用。方法:采用浓度梯度倍增法诱导建立耐药细胞A2780/DXR,并与亲代细胞A2780进行耐药性比较。将耐药细胞A2780/DXR和亲代细胞A2780分别分为空白对照组、VRP逆转对照组(VRP10μmol/ml)、DHA10μmol/ml组、DHA20μmol/ml组、EPA10μmol/ml组、EPA20μmol/ml组,用动物源性ω-3多不饱和脂肪酸对A2780/DXR进行逆转耐药。结果:亲代细胞A2780组,DHA20μmol/ml和EPA20μmol/ml的逆转效果与VRP10μmol/ml的逆转效果具有显著性差异(P<0.05);耐药细胞A2780/DXR组,DHA20μmol/ml的逆转效果与VRP10μmol/ml的逆转效果具有显著性差异(P<0.05)。结论:动物源性ω-3多不饱和脂肪酸DHA、EPA均能有效逆转耐药株对DXR的敏感性,且有明显的剂量依赖性。其中尤以EPA(20μmol/ml)的逆转作用更加明显。Objective: To approach the drug resistance reversion effect of two kinds of animal origin ω -3 polyunsaturated fatty acids, to human ovarian carcinoma cell line A2780 holding doxorubicin (DXR) resistance, by the vitro cytotoxicity experiment of human ovarian cancer cell and doxorubicin persister. Methods :To induce and set the drug resistance cell A2780/DXR by concentration gradient multiplication method, and do drug resistance comparison with parent cell A2780. The drug resistance cell A2780/DXR and parent cell A2780 were divided respectively into blank control group, VRP reverse control group ( VRP 10μmol/ ml), DHA 10μmol/ml group, DHA 20μmol/ml group, EPA 10μmol/rrd group, EPA 20μmol/ml group. To reverse the drug resistance of A2780/DXR by animal origin ω-3 polyunsaturated fatty acids. Results: In parent cell A2780 group, the reverse effect of DHA 20μmol/ml and EPA 20μmol/ml had significant difference (P 〈 0.05) , compared with VRP 10μmol/ml. In the drug resistance cell A2780/DXR group, the reverse effect of DHA 20μmol/ml had significant difference (P 〈 0.05) , compared with VRP 10μmol/ml. Conclusion: Animal origin ω -3 polyunsaturated fatty acids DHA and EPA both reverse effectively the sensitivity of persister to DXR. Both have conspicuous dose dependent. Especially, the reverse effect of EPA 20μmol/ml is more conspicuous.
