摘  要:目的采用合成的人视网膜S-Ag多肽片断第35段抗原决定簇(HS-AgP35)在Lewis大鼠建立实验性葡萄膜炎(EAU)模型。方法雌性Lewis大鼠30只,随机分成3组,每组10只。两实验组中HS-AgP35和BS-Ag分别与CFA混合,致敏Lewis大鼠,正常对照组不做处理。观察大鼠眼部体征和组织病理学改变;ELISA方法检测大鼠血清中抗HS-AgP35/BS-Ag抗体水平;RT-PCR方法检测大鼠脾组织内IL-4和IFN-γ mRNA表达水平。结果两种抗原均能致Lewis大鼠发生EAU,HS-AgP35组100%发病,BS-Ag组60%发病,两者比较差异无统计学意义(P>0.05);HS-AgP35组大鼠发病早(P<0.01),眼部体征和组织病理学改变严重(P<0.01),持续时间长(P<0.01)。两实验组大鼠血清中抗BS-Ag/HS-AgP35抗体水平均高于正常对照组(P<0.01),HS-AgP35组高于BS-Ag组(P<0.05)。两实验组大鼠脾组织中IFN-γ mRNA表达水平均高于正常对照组(P<0.01);HS-AgP35组IL-4 mRNA表达较高(P<0.01),BS-Ag组无明显变化(P>0.05)。结论抗原特异性高的HS-AgP35能诱导Lewis大鼠发生EAU,可建立具有典型眼部表现和组织病理学改变的动物模型。Objective To study the mechanism and further management of autoimmune uveoretinitis(EAU) is an important topic in clinic,and establishment of EAU experimental model is helpful for the understanding and investigation of human autoimmune uveoretinitis.Our study aimed to induce the animal model of EAU with human soluble antigen peptide 35(HS-AgP35). Methods Twenty Lewis rats were immunized with bovine retina S-Ag(BS-Ag) or HS-AgP35 emulsified with equal volume complete Freund’s adjuvant (CFA) respectively and 10 for each,and 10 normal Lewis rats were as control.Clinical signs were monitored daily and histopathological examination was performed under the slim-lamp and light microcope.The antibodies against retinal BS-Ag and HS-AgP35 in animal serum were detected using ELISA,and IL- 4 mRNA and IFN-γ mRNA expressions in the spleens of all the rats were analyzed by RT-PCR. Results Both BS-Ag and HS-AgP35 antigens could induce the uveitis in Lewis rats successfully.The incidence of the rats immunized with HS-AgP35 was 100% and that with BS-Ag was 60%.The onset of EAU was earlier,and the lasting time was longer,and the inflammation was much worse in HS-AgP35 group than BS-Ag group (P〈0.01).There was a significant difference in anti-BS-Ag/HS-AgP35 antibodies level of serum and spleen IFN-γ mRNA expression among all groups(P〈0.01).The spleen IL- 4 mRNA expression level in HS-AgP35 group was higher than orther groups(P〈0.01),but no statistically significant difference was seen between the BS-Ag group and the control group(P〉0.05). Conclusion HS-AgP35-mediated EAU has high incidence and shows a stable,typical clinical sign.This EAU model lay a basis for studing the immunologial mechanism of human autoimmunity uveitis.
