摘  要:目的分析硫辛酸对神经细胞老化进程中细胞凋亡的影响。方法以无血清培养液培养大鼠神经母细胞瘤细胞,建立实验性神经细胞老化模型。建模后细胞随机分为实验组和对照组,分别以含100μmol/L硫辛酸无血清培养液和单纯无血清培养液培养,于培养第5、10、15天时,运用MTT法检测细胞存活率,AnnexinV/PI双染色流式细胞术检测细胞凋亡率。结果与对照组比较,培养第5、10、15天时实验组细胞存活率显著升高,分别为(212.48±12.26)%vs(100.00±14.80)%、(200.55±28.33)%vs(75.45±10.79)%和(96.81±14.50)%vs(28.54±3.14)%;细胞凋亡率则明显降低,分别为(8.60±1.96)%vs (21.46±1.08)%、(18.6±0.60)%vs(30.38±3.25)%和(28.20±3.83)%vs(47.87±5.60)%;两组间细胞存活率和凋亡率比较差异均有统计学意义(P<0.01)。结论硫辛酸可抑制神经细胞老化过程中的细胞凋亡,提高细胞存活率。Objective To study the effect of lipoic acid on apoptosis of neurons during experimental neuronal aging. Methods The mouse neuroblastoma cells with serum-free culture were served as experimental neuronal aging models. The cells were randomly divided into control group ( serum-free culture) and experimental group ( 100μmol/L lipoic acid in serum-free culture). After cultivating for 5, 10 and 15 d, the cell survival rates were detected by MTT method, and the cell apoptosis rates were quantitatively determined using FACS with Annexin V/PI double staining. Results Compared with those in control group, the cell survival rates in experimental group significantly increased[ (212.48 ± 12.26)% vs (100. 0± 14.80) %, (200.55 ± 28.33 ) % vs ( 75.45 ± 10.79) % and (96.81 + 14.50) % vs (28.54 ± 3.14) % , respectively for 5, 10 and 15 d after cultivation] (P 〈0.01 ). Meanwhile, the cell apoptosis rates in experimental group significantly decreased compared with those in control group[ (8.60 ±1.96)% vs(21.46 ±1.08)% , (18.6 ±0.60)% vs(30.38 ±3.25)% and (28.20±3.83) % vs (47.87± 5.60 ) % , respectively for 5, 10 and 15 d after cultivation ] ( P 〈 0.01). Conclusion Lipoic acid can inhibit apoptosis of neurons during neuronal aging and increase cell survival rate.
