葛根素和葛根粗提物对H446细胞生长及对PCNA基因表达的影响

摘　　要：目的:比较葛根素(standard of purepuerarin,SP)和葛根粗提物(pueraria crude extract,CP)对小细胞肺癌H446生长的影响.方法:运用MTT法、流式细胞术、细胞免疫化学方法观察SP和CP对H446细胞生长、凋亡、细胞周期分布以及对PCNA基因表达的影响.结果:CP组和SP组细胞凋亡率分别为14.71%和2.61%,差异具有统计学意义(P<0.05);G0/G1期细胞构成比分别为79.20%和72.20%,G2/M期细胞构成比分别为8.94%和10.50%,与对照组相比,差异均具有统计学意义(P<0.05);CP作用于H446细胞后,PCNA蛋白表达量减少,与对照组相比,差异有统计学意义(P<0.05).结论:CP和SP均可抑制H446细胞增殖,且CP强于SP;SP和CP可诱导细胞凋亡,其机制可能与其导致细胞G0/G1期阻滞、PCNA表达下调有关.AIM： To compare the influence of standard of pure puerarin（SP） and pueraria erode extract （CP） on H446 cell growth. METHODS ： MTT assay, flow eytometry, and immunohistochemistry were used to research the cell proliferation, apoptosis rate and cell cycle distribution and the expression of PCNA gene. RESULTS： The apoptosis rate induced by CP and SP was 14.71% and 2.61% respectively, with significant difference between the two groups（P 〈 0.05 ）. The percentage of G0/Gl cells was 79.20% and 72.20% , and that of G2/M cells 8.94% and 10.50% respectively. Compared with control group, there was significant difference（ P 〈 0.05 ）. The expression of PCNA protein was decreased following CP treatment. CONCLUSION： SP and CP inhibit H446 cell proliferation. The inhibiting effect of CP is stronger than that of SP. SP and CP induce H446 cell apoptosis through blocking cells at G0/G1 stage and down-regulation of PCNA expression.
