缬沙坦治疗免疫性肝纤维化大鼠及其对Bcl-2/Bax表达的影响

摘　　要：目的评价缬沙坦对免疫性大鼠肝纤维化的治疗作用,并观察其对免疫性肝纤维化大鼠肝组织中凋亡相关因子Bcl-2/Bax表达的影响。方法36只雄性SD大鼠随机分为空白对照组、模型组和治疗组。治疗组在造模第6周开始予以缬沙坦干预;第10周实验结束后,观察三组在肝组织纤维化程度、血清透明质酸(HA)、羟脯氨酸(Hyp)水平的变化,免疫组化检测α平滑肌肌动蛋白(α-SMA)、Bcl-2、Bax的表达。结果治疗组大鼠肝纤维化程度、炎症活动度均较模型组明显减轻。模型组和治疗组肝胶原面积分别为(12.5±2.15)%和(5.21±1.21)%(P<0.05)。与模型组比较,治疗组血清HA、Hyp水平均降低(P<0.05)。模型组肝组织α-SMA、Bcl-2和Bax的表达较空白对照组明显增强(P<0.05);而治疗组α-SMA表达明显低于模型组(P<0.05)。治疗组Bcl-2表达较模型组显著降低(P<0.05),Bcl-2/Bax比值降低(P<0.05)。结论缬沙坦能明显减轻猪血清诱导的肝纤维化大鼠的肝纤维化程度、炎症活动度,其机制可能与下调Bcl-2的表达及降低Bcl-2/Bax比值有关。Objective To study the effects of valsartan on immunodamaged hepatic fibrosis and the ratio of Bcl-2/Bax in rats. Methods Thirty-six SD rats were randomly divided into normal control group（N）, model group （M） and drug-treated group（D）. Experimental models were produced by repeated injections of pig serum. The rats in group D were treated with valsartan（30 mg·kg^-1·d^-1 ） from the sixth week. The hepatic fibrosis was evaluated and serum levels of hyaluronic acid（HA） and hydroxyproline（Hyp） were detected at the tenth week in the three groups. The expressions of α-SMA, Bax and Bcl-2 were measured by immunochemical technique. Results The hepatic fibrosis and inflammation were significantly less in group D than those in group M. The area of collagen in group M and D were （12.5±2.15）% and （5.21±1.21）% respectively （P〈0. 05）. The serum HA,and Hyp were significantly lower than those in group M. The expressions of α-SMA,Bcl-2 and Bax were significantly higher in group M than those in group N （P〈0.05）. Compaered with group M, the expression of Bcl-2 was apparantly reduced with a reduced ratio of Bcl-2/Bax in group D （P〈0.05）. Conclusion Valsartan significantly delayed the progression of pig serum-induced liver fibrosis in rats, which may be associated with reduced Bcl-2 expression and the ratio of Bcl-2/Bax.
