摘  要:为了了解光动力学治疗与各种化疗药物合用能否产生协同/拮抗效应,为肿瘤临床提供有用资料,本研究以新型光敏剂叶绿素衍生物CPD4和临床常用的各类化疗药为材料,以人直肠腺癌细胞系HR8348细胞为研究对象,用MTT比色法判别光敏化疗协同效应的结果,对光动力学疗法和各类化疗药的协同/拮抗作用进行系统研究。结果表明:顺铂能明显增强CPD4的光敏杀伤效应,阿霉素(或表阿霉素)在某一剂量范围内亦能增强光敏效应,丝裂霉素有明显的光敏化疗拮抗作用,而氟脲嘧啶、长春新碱与CPD4的光敏效应无协同或拮抗作用。Photodynamic therapy (PDT), the combination of systematically administered photosensitizer and local irradiation with visible light, is a promising new approach for the treatment of cancer. Recently, chrolophyll derivative(CPD 4) has been studied clinically and experimentally and showed a high photodynamic effect. In the present study, the interactions of PDT with newly developed photosensitizer CPD 4 from Chinese Medicine Cansha (Silkworm escrement) and several chemotherapeutic drugs on human rectal cancer cell line HR8348 was investigated. The MTT rapid colorimetric assay was used as measuring indicator of photodynamic cytotoxicity. The results showed that cisplatin markedly increased the photodynamic effect of CPD 4; under given drugs dose, adrimycin/epirubisin could increase the effect; mitomycin C could decrease the efficacy of photodynamic inactivation of CPD 4; 5 fluorouracil and vincristin had no interaction with PDT of CPD 4.
