摘  要:目的探讨角质生长因子(KGF)是否具有调节正常滋养细胞及绒癌细胞系JEG-3、BeWo的增殖和分泌HCG的功能,以及彼此间有无差异。方法采用MTT法、IEMA法测定早孕绒毛膜滋养层细胞和绒癌细胞系JEG-3、BeWo的增殖和分泌HCG能力。结果KGF对滋养细胞及JEG-3、BeWo细胞增殖均有促进作用,滋养细胞存在最大作用浓度,在100ng/ml以上,生长曲线受到抑制。而对JEG-3细胞,有剂量依赖效应,随浓度加大,增殖能力增强。KGF对BeWo同样具有促进作用,类似JEG-3,无统计学意义。KGF对上述细胞分泌HCG的影响与生长曲线基本相一致。KGF促JEG-3、BeWo的增殖和分泌能力明显高于滋养细胞(P<0.01)。结论KGF参与早孕期滋养细胞的增殖及分泌,但受体内母胎界面内环境调控。同时KGF能明显增强肿瘤细胞JEG-3、BeWo的无节制增殖、分泌过程。提示KGF可能参与滋养细胞肿瘤的发生、发展过程。Objective To investigate the relationship of the proliferation and secretion function between trophoblast JEG-3, BeWo and KGF in vitro. Method Normal placenta trophoblast tissues were collected by suction curattage from maternity ward.The villous tissues were minced into small pieces and incubated in DEME medium. All tissues were passage by trypsinization. Trophoblast, JEG-3 and BeWo were cultured with KGF for 48 hrs.The collected culture medium were stored at -20℃ for IEMA.MTT assay and IEMA method were employed for observing the proliferation and secretion of these cells. Result KGF can stimulate the proliferation of trophoblast, BeWo and JEG-3, and plated a dural role in trophoblast growth and secretion. KGF stimulated proliferation and HCG secretion, but inhibited these function by the highest dose (100ng/ml).Otherwise, a significant amplified effect were noted on cell proliferation of BeWo and JEG-3 as we 11 as on secretion at the highest concentration(100ng/ml).The proliferation of BeWo was lower than that in JEG-3. At the same time, the effect of KGF in ehoriocarnoma cell lines were significantly higher than that in trophoblast (P〈0.01, P〈0.01 ). Conclusion KGF plays different role in proliferation and secretion of HCG between trophobalst and choriocarnoma cells. These different functions between trophoblat and choriocarnoma cells suggest that KGF may play a key role in the pathogenesis of GTD.
