Analysis on Concentration of Amino Acids in the Hippocampus of Status Epilepticus Rats with Transplantation of Human Mensenchymal Stem Cells

摘  要:目的了解人骨髓间充质干细胞(hMSC)移植后致痫大鼠海马组织中氨基酸含量变化。方法采用氯化锂-毛果芸香碱诱发雄性SD大鼠癫痫持续状态(SE)模型。大鼠随机分为MSC组、磷酸缓冲液(PBS)组、假手术组(Sham)和正常对照组。术后10周断头取海马组织,测定兴奋性神经递质谷氨酸(Glu)、天冬氨酸(Asp)及抑制性神经递质γ-氨基丁酸(GABA)、甘氨酸(Gly)含量。结果hMSC组和正常对照组海马组织Asp、Glu和GABA含量差异有统计学意义(P<0.05),Gly含量差异无统计学意义(P=0.0617);致痫成功的hMSC组、PBS组和假手术组海马组织中氨基酸含量比较差异均无统计学意义。结论所有致痫大鼠海马组织中氨基酸含量均低于正常对照组,hMSC移植并未明显改变SE大鼠海马组织中氨基酸含量。Objective To observe the concentration changes of amino acids in the hippocampus of status epilepticus(SE) rats with transplantation of human mensenchymal stem cells(hMSC). Methods The SE rat models were induced by Lithium-pilocarpine in DS rats. The SD rats were randomly divided into four groups: hMSC group, phosphate buffered saline(PBS) group, sham group and normal control group. After 10 weeks, all rats hippocampus tissue was taken out and Glu, Asp, GABA and Gly in the hipocampus of the samples were processed and determined by means of high performance liquid chromatography. Results The concentrations of Glu, Asp, GABA in the hippocampus between hMSC group and normal control group were significantly different (P〈0.05). The concentration of Gly was lower in hMSC group(P=0. 0617). The concentrations of Glu, Asp, GABA and Gly in the hippocampus among hMSC group, PBS group and sham group were not significantly different. Conclusions The concentrations of amino acids in the hippocampus of SE rats were lower than that in normal control group. The transplantation of hMSC might not change the concentrations of amino acids obviously in the hippocampus.
