摘  要:目的:探讨大鼠局灶性脑缺血后神经元凋亡的细胞周期特异性分布以及相关基因表达之间的内在关系。方法:建立大鼠大脑中动脉阻塞模型,利用流式细胞分选和原位末端标记及RT-PCR技术,检测正常组和缺血后大鼠脑组织中的神经元凋亡的细胞周期特征,及其Cyclin D1和Cyclin B1基因的表达规律。结果:神经元凋亡可以发生在细胞周期的各个时相,细胞凋亡多发生在G0/G1期,而且神经元也可在S期甚至到达G2期发生凋亡。正常脑组织中神经元表达Cyclin D1,缺血后表达增加;Cyclin B1在正常脑组织的神经元中不表达,缺血后呈现弱表达。结论:脑缺血后神经元可以重新进入细胞周期,其细胞凋亡是多点启动的,可发生在细胞周期的各个时相。Cyclin D1和Cyclin B1的异常激活可能参与了缺血后神经元凋亡的细胞周期调控过程。Objective: To investigate the relationship between the expression of Cyclin D1, Cyclin B1 and cell cycle specific apoptosis of neurons after focal cerebral ischemia in rats. Methods: Ischemia/reperfusion injury was induced by temporary middle cerebral artery occlusion(MCAO). And the brain was taken at 3 days after injury, then analyzed the cell cycle specificity of apoptosis and apoptosis rate by flow cytometry combined with ter- minal deoxynucleotidyl transferase mediated dUTP nick end labeling(TdT) methods. Andthe expression of Cyclin D1, Cyclin B1 mRNA were detected by RT - PCR. Results: The apoptotic neurons after MCAO occurred in each phase of cell cycle, the most were in GjG1 phase, and neurons may re - entry S phase and G2 phase. Cyclin D1 was found in neurons of normal group and its expression increased after MCAO, and in normal neurons Cyclin B1 was not found but was detected mildly in neurons following MCAO. Conclusion: Neurons may reentry cell cycle and its apoptosis may occur in each phase, which was induced at different check- point. It was suggested that the abnormal expression of Cyclin D1 and Cyclin B1 may participate in the process of neuron apoptosis after focal cerebral ischemia.
