幽门螺杆菌阳性与消化性溃疡患者IL-8、IL-10、IL-12及NF-κB表达的关系研究

摘　　要：目的研究幽门螺杆菌(H.pylori)与消化性溃疡(PU)患者胃黏膜IL-8、IL-10、IL-12及NF-κB表达的关系。方法采用荧光定量PCR法测定168例H.pylori阳性和46例H.pylori阴性的消化性溃疡患者以及30例正常对照者的胃黏膜IL-8、IL-10、IL-12及NF-κB mRNA含量,H.pylori阳性患者清除H.pylori后复查。应用免疫组化检测IL-8、IL-10、IL-12与NF-κB在胃黏膜组织的表达。结果H.pylori阳性组胃黏膜IL-8I、L-12及NF-κBmRNA含量较H.pylori阴性组和正常对照组明显增高(P均=0.000);H.pylori阳性患者清除H.pylori后胃黏膜IL-8、IL-12及NF-κB mRNA含量降低(P均<0.05)。免疫组化结果显示H.pylori阳性组较H.pylori阴性组以及正常对照组IL-8、IL-12、NF-κB表达明显增强(P均<0.05)。结论H.pylori感染可以诱导胃黏膜合成和释放IL-8、IL-12及NF-κB,它们是引起PU炎症以及进一步病理损害的重要因子。Objective It is to study the relation between Helicobacter pylori （H. pylori） and expression of IL- 8, IL- 10, IL- 12 and NF- κB in gastric mucosa of patients with peptic ulcer （PU）. Methods The contents of IL- 8, IL- 10, IL- 12 and NF - κB in gastric mucosa of 168 PU patients with H. pylori positive and 46 PU patients with H. pylori negative and 30 normal control people were determined with fluorescence quantitative PCR method. H. pylori positive patients were rechecked after H. pylori eradication. The expressions of IL - 8, IL - 10, IL - 12 and NF - κB in gastric mucosa were detected with immunohistochemisty. Results The contents of IL - 8, IL - 12 and NF -κB mRNA in H. pylori positive group were significantly higher than those in H. pylori negative group and normal control group （P = 0. 000）, their IL- 8, IL- 12 and NF-κB mRNA were decreased after H. pylori eradication （P〈0.05）. Immunohistochemical results showed that the expressions of IL - 8, IL - 12 and NF - κB in H. pylori positive group increased obviously than H. pylori negative group and normal control group （ P 〈 0.05 ）. Conclusion H. pytori infection can induce gastric mucosa synthesizing or releasing IL - 8, IL - 12 and NF- κB which may be important factor to induce inflammation and progressive pathological damage.
