近交系豫医无毛小鼠微卫星DNA多态性分析

摘  要:目的:研究微卫星DNA多态性在豫医无毛小鼠等近交系小鼠遗传监测中的应用。方法:选取小鼠不同染色体上的18个微卫星位点(D3Mit17、D3Mit18、D3Mit15、D3Mit21、D3Mit22、D6Mit192、D6Mit81、D6Mit36、D6Mit149、D16Mit4、D7Nds1、D2Nds3、D3Mit16、D3Mit85、D3Mit41、D4Mit2、D5Mit66、D7Mit164),应用PCR技术对7个近交系小鼠(豫医无毛小鼠、BALB/C、C57、CBA、DBA/2、C3H/HeJ、FVB/NJ)进行了微卫星多态性分析。结果:18个微卫星DNA具有稳定扩增效果。不同品系个体之间11个位点(D3Mit17、D3Mit18、D3Mit15、D3Mit21、D3Mit22、D6Mit192、D6Mit81、D6Mit36、D6Mit149、D16Mit4、D7Nds1)具有多态性,其中5个位点(D6Mit36、D6Mit81、D16Mit4、D6Mit149、D7Nds1)具有显著多态性;同一品系不同个体之间表现为单态性。结论:所检测的小鼠符合近交要求,运用筛选出的11个位点(D3Mit17、D3Mit18、D3Mit15、D3Mit21、D3Mit22、D6Mit192、D6Mit81、D6Mit36、D6Mit149、D16Mit4、D7Nds1)进行微卫星多态性分析,能够快速、经济地对近交系小鼠进行品系鉴别和遗传背景监测。同时也反映了近交系豫医无毛小鼠与其他品系小鼠之间亲缘关系的远近。Aim: To analyse microsatellite DNA loci polymorphisms and genetic monitoring of Yuyi hairless mice and other inbred mice strain.Methods:A total of 18 microsatellites DNA loci(D3Mit17,D3Mit18,D3Mit15,D3Mit21,D3Mit22,D6Mit192,D6Mit81,D6Mit36,D6Mit149,D16Mit4,D7Nds1,D2Nds3,D3Mit16,D3Mit85,D3Mit41,D4Mit2,D5Mit66,D7Mit164)on different chromosomes in seven inbred mice strains(Yuyi hairless mice,BALB/C,C57,CBA,DBA/2,C3H/HeJ,FVB/NJ) were investigated by PCR analysis.Results:A total of 18 microsatellite DNAcould be amplified efficiently.And 11 loci(D3Mit17,D3Mit18,D3Mit15,D3Mit21,D3Mit22,D6Mit192,D6Mit81,D6Mit36,D6Mit149,D16Mit4,D7Nds1)displayed polymorphisms among the various strains,among which 5 loci(D6Mit36,D6Mit81,D16Mit4,D6Mit149,D7Nds1)displayed remarkable polymorphism,and no polymorphism in the same mice strains.Conclusion:These tested mouse have accorded with the requests of inbred.Analysis of microsatellites DNA polymorphisms by the 11 microsatellites DNA(D3Mit17,D3Mit18,D3Mit15,D3Mit21,D3Mit22,D6Mit192,D6Mit81,D6Mit36,D6Mit149,D16Mit4,D7Nds1)is a fast and economical way for strain identification and genetic monitoring among inbred mouse strain.And the research reflects their ancestors′ kindred relationship of Yuyi hairless mice and other strain mice.
