摘  要:建立了柱前衍生反相高效液相色谱-紫外检测富硒脱水菜心中硒代胱氨酸(SeCys)、硒甲基半胱氨酸(SeMeCys)和硒代蛋氨酸(SeMet)的方法,优化了样品预处理方法,并评价其定量分析的线性、准确性、重复性和检出限,结果显示:以0.1mol/LHCl为提取溶剂,超声处理8h,提取效果最佳;3种硒代氨基酸的平均加标回收率为95.7%~112.9%;在各硒代氨基酸的线性范围内,线性关系良好(R>0.999);保留时间和峰面积重复性的相对标准偏差(RSD)均分别低于2.0%和5.0%;酸性条件下的检出限为0.004~0.009mg/L。A reversed phase high performance liquid chromatography/variable wavelength detector with precolumn derivatization was developed for the simultaneous determination of selenocystine(SeCys), seleno-methylcysteine(SeMeCys) and selenomethionine(SeMet). This method was validated for linearity, accuracy, repeatability and limits of detection (LOD). The results showed that average recoveries of seleno-amino acids standards ranged from 95.7% to 112.9%, linearity was good (R^2〉0.999); the relative standard deviations of repeatability for retention time and peak area were less than 2. 0% and 5.0%, respectively; LOD was 0. 004~0. 009 mg/L in acid condition. The method is simple, accurate, and sensitive and reliable for quality control of enriched--selenium dehydrated Brassica Chinensis.
