摘  要:背景 WNK1是新近发现的肾脏多个离子转运体及通道的调控蛋白,与醛固酮离子转运途径有关。目的探讨高钾饮食致体内醛固酮增高对大鼠肾脏 WNK1不同转录本表达的作用及其意义。方法采用高钾饮水饲养 Wistar 大鼠8只,检测血清、24 h 尿液离子质量浓度及血清醛固酮质量浓度。半定量 RT-PCR 方法检测大鼠肾脏 WNK1-L和 WNK1-KS 基因表达情况。结果与正常对照组(n=8)比较,高钾组(n=8)血清醛固酮水平显著升高[(1.86±0.66)比对照组:(0.25±0.04)μg/L,P<0.01],为对照的7.4倍。血清及尿液钾离子和尿液氯离子质量浓度升高(P<0.05),尿液钠离子质量浓度下降(P<0.05),血清钠离子和氯离子无差异(P>0.05)。高钾组WNK1-L 表达量无差异(0.27±0.07比0.26±0.07,P>0.05),WNK1-KS 表达量显著上升(1.60±0.22比0.38±0.08,P<0.01)。结论高钾饮食导致大鼠醛固酮质量浓度升高,醛固酮可通过调控 WNK1-KS 表达起到排钾保钠的生理作用。Background WNK1 is a recently found upstream kinase of multiple transporters and channels in kidney, while might be involved in the biological effect of aldosterone. Objective To investigate the effect of increases in aldosterone induced by high dietary potassium intake on the expression of rat kidney WNK1 isoforms. Methods High KC1 solution(0.5 mol/L) in diet was administrated to Wistar rats(n=8). The concentrations of electrolytes and aldosterone in serum as well as 24 hour electrolytes concentrations in urine were measured. RT-PCR was used to detected the expression of rat kidney WNK1 isoforms. Results Serum concentrations of aldosterone in high K^+ diet rats was 7.4 times of that in the control group(1.86±0.66 vs control:0.25±0.04μg/L, P〈0.01 ). Serum K^+ and urine K^+ and Cl^- in the experimental rats were significant higher than those in normal rats while the urine Na^+ was lower( P〈0.05 ). There were no significant difference of serum Na^+ or Cl^- between two groups(P〈0.05). The expression of WNK1-KS is higher in the experimental rats than that in control rats(1.60±0.22 vs 0. 38±0. 08, P〈0.01) while no differences was found in the expression of WNK1-L in both groups (0.27±0.07 vs 0.26±0. 07, P〉0.05). Conclusion The high dietary potassium intake induces the secretion of aldosterone. Aldosterone regulating serum and urine sodium and potassium is associated with upregulated expression of WNK1-KS.
