摘  要:目的:研究葶苈子水提液对压力负荷大鼠心肌CYP11B1、CYP11B2及TGF-β1 mRNA表达的影响。方法:腹主动脉不完全结扎法(AAB)制作大鼠心肌肥厚、心室重构模型。药物干预30d后,称重法测定心脏指数(HW/BW)、左室重量指数(LVW/BW),Real-Time PCR反应技术,以Rat 18S为内参照,测定心肌CYP11B1,CYP11B2,TGF-β1 mRNA的表达水平。结果:模型组大鼠心脏指数增高,醛固酮合成基因和转化生长因子β1表达增强。葶苈子水提液能改善心肌肥厚指数,降低大鼠心肌醛固酮合成基因CYP11B1、CYP11B2及TGF-β1的mRNA表达水平。结论:葶苈子水提液可能通过抑制心肌醛固酮合成相关基因CYP11B1、CYP11B2 mRNA表达水平,降低转化生长因子β1的合成而抑制心室重构的发生。Objective:To investigate the effects of Semen descurainiae and Captopril on CYP11B1, CYP11 B2 and TGF-β1 mRNA expression of heart tissue in rats treated with Abdominal Aortic Banding. Methods:Ventrieular remodeling was induced by abdominal aortie banding (AAB) in rats. Aider 30 days' treatment,the ratios of LVW/BW (left ventricle weight/body weight),HW/BW (heart weight/ body weight) were calculated ;Then the CYP11 B1 ,CYP11 B2 and TGF-β1 mRNA expression of left ventricle were detected by Real-time PCR ,respeetively . Results :The experimental data demonstrated that Semen descurainiae decreased the indexes of LVW/BW and HW/BW ,down-regulated CYP11B1 ,CYP11B2 and TGF-β1 mRNA expression in left ventricle ( P 〈 0. 05 ). Conclusion: Semen descurainiae can significantly inhibit the experimental ventricular remodeling; the mechanism is related to its ability to attenuate the mRNA expression of CYP11B1 ,CYP11B2 and TGF-β1 in left ventricle. The inhibition of aldosterone key gene expression by Semen descurainiae may contribute to its effect on restraint cardiac remodeling.
