摘 要:采用胶束毛细管电泳在线扫集技术分离测定了中药穿心莲中脱水穿心莲内酯和穿心莲内酯:电泳条件:以20mmol/LH3BO3—10mmol/LNaH2PO4—50mmol/LSDS(含体积分数20%甲醇,pH2.4)为电泳运行缓冲溶液,未涂层石英毛细管(58cm×50um i.d.,有效长度为41.2cm)为分离通道,重力进样,进样高度为11cm,-20kV恒压,检测波长为246nm。富集倍数可以达到200倍以上。在5.70~91.20mg/L,和3.96~31.68mg/L范围内呈良好的线性关系,对两种内酯分别进行了定量分析。加标平均回收率脱水穿心莲内酯为100.80%,穿心莲内酯为98.06%。Andrographis paniculata nees is one of the well-known traditional chinese medicinal materials, in which andrographolide and dehydroandrographolide are two major active compounds. In this work, a sweeping micellar electrokinetic capillary chromatographic method has been developed for separation and quantitative analysis of these two compounds in andrographis paniculata nees. Electrophoretic conditions are as follows: uncoated fused silica capillary (41.2 cm/58 cm, 50 tan i. d. ), 20 mmol/L boric acid-10 mmol/L sodium dodecylsulfate-50 mmol/L SDS containing 20% methanol ( V/V) at pH 2.4, applied voltage - 20 kV, detection wave-length 240 nm, injection by gravity with the height 11 era. Up to 200-fold improvement in enrichment was achieved by using this sweeping injection technique. Both of the analytes were separated successfully within 10 min under the optimum conditions. The relative peak areas of the two analytes increase linearly with the increase of their concentrations in the range of 5.70 - 91.2 mg/L for dehydroandrographolide and 3.96 - 31.68 mg/L for andrographolide, and the average recoveries are 100.8% and 98.06%, respectively.
