摘  要:目的探讨多药耐药相关蛋白(multidrug resistance-associated,MRP)基因表达与卵巢癌临床病理组织学特征的相关性。方法应用半定量逆转录-聚合酶链反应(reverse transcription-polymerase chain reaction,RT-PCR)技术,检测了37份原发上皮性卵巢癌(primary epithelial ovarian cancer,PEOC)组织、10份正常卵巢组织和20份正常女性外周血单核细胞中MRP基因表达情况。结果PEOC中MRP基因表达水平明显高于正常卵巢组织(P<0.05),与外周血单核细胞比较差异无统计学意义(P>0.05);MRP基因表达与PEOC患者临床期别及病理组织学类型无关(P>0.05),但随病理组织学级别升高而升高(P<0.05);MRP和MDR1基因复合表达与患者无进展生存期有关(P=0.005),但与总体生存期无关。结论在组织细胞中MRP基因具有广泛性表达,与PEOC的分化程度密切相关,是其固有耐药的重要原因之一,对患者临床疗效判断和预后预测有一定价值。Objective To explore the association between the expression of multi-drug resistance-associated protein(MRP) and the elinicopathological features of the primary epithelial ovarian cancer( PEOC), and to investigate the clinical significance of the expression of MRP gene in PEOC. Methods Using the reverse transcfiption-polymerase chain reaetion(PT-PCR) assay,the expressions of MRP gene in 37 PEOC, 10 normal ovary tissues and 20 peripheral blood monouclear cells(PBMC) samples were examined. Results The expressions of MRP gene in PEOC were higher than those in ovarian tissues ( P 〈 0. 05 ). But compared with PBMC, there were no differences( P 〉 0. 05 ). There were no correlations respectively between MRP gene expressions and FIGO stage or histological type of PEOC. But the poorly the histological differentiation was,the higher the expression of MRP gene was. Patients with MRP gene high expression tumors had a significantly shorter progression-free survival than those with MRP gene low expression. Conclusions MRP gene expression related to resistance of PEOC are major causes of resistance to MDR related drug. Detecting the expression of MRP gene can help in selecting chemotherapeutic drugs and predicting the prognoses of patients.
