食管鳞癌组织中HIF-lα、Caspase-3蛋白的表达变化及意义

摘　　要：目的探讨食管鳞癌组织中缺氧诱导因子(HIF)-1α、半胱氨酸天冬氨酸蛋白酶(Caspase)-3蛋白的表达变化及其与肿瘤浸润、转移的关系。方法应用免疫组化SP法检测49例食管鳞癌中HIF-1,αCaspase-3蛋白的表达。结果食管鳞癌组织中HIF-1α、Caspase-3蛋白阳性表达率分别为51.0%(25/49)、34.7%(17/49),与阴性对照组的0、100%相比,P均<0.05。HIF-1α、Caspase-3蛋白表达与食管鳞癌组织浸润深度及有无淋巴结转移有关(P均<0.05),与分化程度无关(P>0.05)。HIF-1α、Caspase-3蛋白在食管鳞癌中的表达呈负相关(P<0.05)。结论食管鳞癌组织中存在HIF-1α蛋白的高表达和Caspase-3蛋白的缺失或低表达。这一变化与食管鳞癌的浸润转移有关,其机制可能与HIF-1α蛋白下调Caspase-3蛋白的表达有关。Objective To indentify the expression of HIF-lα and Caspase-3 proteins, and investigate the relationship between them and the invasion and metastasis of esophageal squamous cell carcinoma. Methods The expressions of HIF-lα and Caspase- 3 protein were detected in 49 cases of esophageal squamous cell carcinoma by streptomycete avidin peroxides（SP） immunohistochemistry. Results The positive rates of HIF-lα and Caspase-3 protein in esophageal squamous cell carcinoma tissue were 51.0% （25/49） ,34.7% （ 17/49）, compared with control groups（0,100% ）, P〈 0.05. The expression of HIF-lα and Caspase-3 protein were correlated with infiltration depth and lymph node metastasis（ P 〈 0.05）, but not related with cell differentiation grade（ P 〉 0.05）. The expression of HIF-lα and Caspase-3 protein in esophageal squamous cell carcinoma tissue had inverse correlation（ P 〈 0.05）. Conclution There is HIF-lα protein over expression and Caspase-3 protein loss or decreased expression in esophageal squamous cell carcinoma tissue. The mechanism may be that the expression of HW-lα protein can decrease the expression of Caspase-3 protein.
