摘 要:目的用实时荧光定量PCR的方法探讨miRNA-142-5p在肾透明细胞癌中的表达,为进一步研究其功能做准备。方法提取30例手术患者肾癌组织和癌旁正常组织中的miRNAs,在基因芯片判断的基础上,用RT-PCR的方法初步检测miRNA-142-5p在肾癌组织和癌旁正常组织中的表达情况,后采用Real-time PCR的方法定量分析miRNA-142-5p在肾癌组织及癌旁正常组织中的表达情况。结果相对于癌旁正常组织,miRNA-142-5p在肾癌组织中高表达,平均表达升高4.13倍。30例标本中有21例肾癌标本中miRNA-142-5p的表达明显高于其在癌旁正常组织中的表达,仅有9例肾癌标本中miRNA-142-5p的表达低于其在癌旁正常组织中的表达。7例G1期的肾癌标本均较对应的癌旁正常组织低表达,而较癌旁正常组织高表达的肾癌组织分布在G2和G3期,初步显示与分期有一定的关系。结论miRNA-142-5p在肾癌组织中显著存在高表达,提示其与肾透明细胞癌的发生与演进有关,有望成为指导肾细胞癌诊断、治疗、预后的新靶点。Objective To quantitatively detect the level of miRNA-142-5p expression in human renal cell carcinoma with the approach of real-time PCR and to prepare for the research of its function. Method The total miRNAs were isolated from human kidney cancer tissues and adjacent normal tissues from 30 patients with renal cell carcinoma who had been operated in the recent one year. Before using the quantitatively real-time PCR to detect the expression of miRNA-142-5p in both normal and abnormal tissues, the RT-PCR was performed firstly based on the result of micro-array analysis. Results miRNA-142-5p expression was higher in tumor tissues than that in paratumor tissues and the mean time was 4. 13 (P 〈 0. 01 ). miRNA-142-5p in 21 samples from 30 patients with renal cell carcinoma showed higher expression than that in normal tissues,while miRNA-142-5p in the other 9 samples showed lower expression than that in normal tissues. 7 samples which were classified as G1 displayed negative results,otherwise,the positive samples all scattered in G2 and G3. Conclusion miRNA-142-5p may be a new targetin diagnos is therapy and prognosis of patients with renal cell carcinoma.
