摘  要:目的:研究并分析新型紫杉醇脂质纳米粒子(paclitaxel nanoparlicle,TAX-NLC)对KB、SKOV3细胞的抑制作用,为研制新型肿瘤靶向治疗药物提供实验依据。方法:激光共聚焦显微镜动态观察TAX-NLC进入细胞的过程,半定量测定TAX-NLC在细胞内的浓度;MTT法测定TAX-NLC对KB、SKOV3细胞增殖的抑制率;锥虫蓝拒染法计数TAX-NLC作用后的活细胞数,绘制细胞生长曲线。结果:激光共聚焦显微镜动态观察发现细胞内荧光强度逐渐增加,约2 h时达到饱和,胞内外荧光强度差达20~40倍;MTT结果表明,TAX-NLC、紫杉醇(paclitaxel,TAX)对细胞的生长均有抑制作用,并且存在时间和剂量的依赖关系,用药后72 h后抑制率最高达92%;细胞生长曲线显示,TAX-NLC、TAX对SKOV3、KB细胞有明显的抑制作用,前者的作用强于后者。结论:TAX-NLC可能是通过主动转运将紫杉醇载带入细胞内,并在细胞内浓聚,对KB、SKOV3细胞的生长具有较强的抑制作用;其可能是一个有前途的新型肿瘤靶向治疗药物。Objective:To study and analysis the inhibitory effects of a new type paclitaxel-nanoparticle (TAX-NLC) on KB and SKOV3 cells and provide experimental evidence for developing a novel tumor-targeted pharmaceutical agent. Methods:The dynamic process of TAX-NLC into tumor cells was observed by laser confocal microscopy and the intracellular concentration of TAX-NLC was determined semi-quantitatively. The inhibitory effects of TAX-NLC on the proliferations of KB and SKOV3 cells were measured by MTT assay. The viable cells were detected by trypan blue staining and the cell growth curve was calculated. Results: Under the laser confocal microscope, the TAX-NLC wrapped FITC entered the cells gradually. The intracellular fluorescence intensity reached the saturation state about two hours later, which was 20-40 times higher than extracellular fluorescence intensity. TAX-NLC and TAX inhibited proliferation of KB or SKOV3 cells in a dose- and time-dependent manner. The maximal inhibitory ratio was 92% after 72 h treatment. From the cell growth curve, we observed that both TAX-NLC and TAX had inhibitory effects on the proliferation of SKOV3 and KB cells, but the inhibitory potency of TAX-NLC was obviously stronger than TAX. Conclusion:TAX-NLC carries TAX into tumor cells via active transportation, accumulates in cytoplasm, and exerts stronger inhibitory effects on the growth of KB and SKOV3 cells. TAX-NLC has the potential to become a novel tumor-targeted therapeutic agent.
