摘  要:目的:研究亚硒酸钠对人结肠癌细胞的抑制机制。方法:通过DNA末端原位标记染色法(TUNEL)检测亚硒酸钠溶液对人结肠癌细胞株SW-111C凋亡的影响;应用免疫组织化学染色技术观察亚硒酸钠对人结肠癌细胞株SW-111C p16,p21,PCNA,CD44基因表达的影响;并测定亚硒酸钠对小鼠T淋巴细胞亚群的影响。结果:TUNEL染色法表明亚硒酸钠作用细胞后凋亡指数明显升高(P<0.01),且具有剂量-效应关系;亚硒酸钠作用后人结肠癌细胞SW-111C p16和p21基因的蛋白表达上调,PCNA和CD44基因的蛋白表达下调;亚硒酸钠能使小鼠CD3+,CD4+细胞数增加,而且CD4+/CD8+比值升高,与对照组比较差异有统计学意义(P<0.05)。结论:亚硒酸钠可通过诱导细胞凋亡,影响p16,p21,PCNA,CD44基因表达和提高机体的免疫功能而抑制SW-111C的生长。Objective: To investigate the inhibition mechanism of sodium selenite on SW-111C cells. Methods: The apoptosis of SW-111C cells treated with sodium selenite was observed by TUNEL. The expression of p16, p21, PCNA, CIM4 treated with sodium selenite was performed by immunohistochemistry technique. The T-lymphocyte subsets were detected by animal experiment. Results: The apoptotic index of SW-111C cells increased and was positively correlated to the concentration of sodium selenite( P 〈 0.01 ). The expression of p16 and p21 was up-regulated and the expression of PCNA and CD44 was down regulated after treated by sodium selenite. After exposure to sodium selenite, the number of CD3^+ and CD4^+ cells in mice increased and CD4^+ /CD8^+ increased too( P 〈 0.05 ). Conclusion: The sodium selenite could inhibit the proliferation of SW-111C cells by inducing cell apoptosis, and influence the expression of p16,p21, PCNA, CD44 with improving immune function.
