摘 要:目的:探讨微管相关蛋白Ⅱ(MAP2)及突触素与癫后海马兴奋性神经网络中苔藓纤维发芽和突触重建的关系。方法:KA诱导的成年大鼠MTLE模型用免疫组织化学法检测癫发作1d,1、2、3、4周大鼠海马内MAP2、突触素的表达。结果:癫组MAP2的表达于癫发作后1周在齿状回分子层、CA3区辐射层已有上升,2周达到高峰,4周恢复接近对照组水平;突触素的表达于发作后1周在齿状回分子层和CA3区辐射层开始上升,2周时达高峰并持续至4周。结论:癫发作后海马齿状回、CA3区MAP2、突触素的表达升高,在时间和部位上的动态变化与已知的癫后MFS和突触重建的改变相一致,提示可能与突触重建有关,可作为癫后海马MFS和突触重建的间接观察指征。Objective:To investigate the relationship between the expression change of microtubule-associated protein Ⅱ (MAP2)and synaptophysin in hippocampus and mossy fiber sprout(MFS) and synaptic reconstruction of excitatory neural network after mesial temporal lobe epilepsy (MTLE). Methods: 40 rats were injected kainic acid through lateral cerebral ventricle to Establish the MTLE model, and 25 rats injected salt solution as controls. Dynamic change of the expression of MAP2 and synaptophysin in hippocampus after epileptic seizure in 1d, 1w, 2w, 3w and 4w by using immunohistochemcal methods. Result: Compared with the controls, the expression of MAP2 in molecular layer of dentate gyrus and stratum radiatum of CA3 region in hippocampus increased in 1w, and rose to the peak in 2w, decreed to the level near to the controls. In contrast with MAP2, the expression of synaptophysin in molecular layer of dentate gyrus and stratum radiatum of CA3 region in hippocampus was enhanced from lw to 4w. Conclusion:The expression of MAP2 and synaptophysin in dentate gyrus and CA3 region of hippocampus enhanced after KA induced epileptic seizure in MTLE model rats. The expression change of MAP2 and synaptophysin revealed in our experiments suggests that MAP2 and synaptophysin may play a role in MFS and synaptic reconstruction after epilepsy.
