摘  要:目的:探讨外源性神经生长因子(NGF)对局灶性脑缺血再灌注大鼠皮质白细胞介素1β(IL-1β)表达的影响。方法:线栓法制作大鼠局灶性脑缺血再灌注模型,应用免疫组织化学显色、免疫印迹分析方法,结合图像分析技术检测大鼠缺血侧顶叶皮质IL-1β蛋白表达变化。结果:假手术组大鼠顶叶皮质IL-1β没有表达,缺血再灌注组顶叶皮质IL-1β蛋白在各时间点明显表达,IL-1β蛋白阳性产物的光密度值高于假手术组;NGF组IL-1β蛋白阳性产物的光密度值低于缺血再灌注组。结论:脑缺血再灌注损伤后诱导IL-1β蛋白表达,NGF明显降低缺血再灌注大鼠顶叶皮质IL-1β蛋白表达,这可能是NGF发挥神经保护作用的分子机制之一。Objective: To investigate the effect of the exogenous nerve growth factor (NGF) on the expressions of IL-1β in the rat parietal cortex following focal cerebral ischemia/reperfusion (I/R). Methods: Middle cerebral artery occlusion (MCAO) was established by occluding the right middle cerebral artery using intraluminal suture method. The expression of IL-1β was determined by SABC immunohistochemical method and Western blot analysis, and the positive products were ana lyzed by image analysis system. Results: The expressions of IL-1β protein in the parietal lobe cortex were absent in the sham group, were obviously expressed at 3 h, reached a maximum at 48 h, and gradually decreased at 72 h in the I/R group. The expressions of IL-1β protein were significantly higher in the I/R group compared with those in the sham group. The expres sions of IL-1β protein were lower in the NGF group than those in the I/R group. Conclusion: IL-1β protein is induced and expressed after I/R in the rat cortex. NGF plays a role in downregulating the expressions of IL-1β protein in the parietal lobe cortex following focal ischemia/reperfusion, which may be involved in the protective mechanism to ischemic neurons.
