摘 要:目的探讨子宫内膜癌Ishikawa细胞系中细胞膜孕激素受体的表达情况。方法荧光显微镜及流式细胞仪检测不同浓度的异硫氰酸荧光标记的牛血清白蛋白-孕酮复合物(P-BSA-FITC)作用下,子宫内膜癌Ishikawa细胞荧光标记情况。结果P-BSA-FITC作用于Ishikawa细胞系后,在细胞膜上可见荧光染色,而核内未见荧光染色。流式细胞仪检测结果显示,25μg/ml组在各时点的荧光标记细胞数明显高于10μg/ml和50μg/ml组(P<0.05),在作用开始时呈时间依赖性,至60min达高峰,继续延长作用时间并不能增加细胞的荧光标记率(P>0.05)。结论在子宫内膜癌Ishikawa细胞系表面存在着孕激素膜受体,其与孕激素的结合能力大小与孕激素的浓度和作用时间有关。Objective To explore the expression of membrane progesterone receptor in endometrial carcinoma. Methods Endomtrial carcinoma cell line Ishikawa was treated by non-permeable progesterone-BSA conjugate labeled with fluorescein isothiocyanate (P-BSA-FITC). The location of P-BSA-FITC in the cells was observed using fluorescence microscopy. The percentage of labeled cells at different concentrations of P-BSA-FITC was determined by flow cytometry. Alternatively, BSA-FITC as positive control group was applied to validate the speci- ficity of P-BSA-FITC location on cell plasma membrane. Results Fluorescence on the membrane was visualized in cells treated with P- BSA-F1TC. When cells were treated with BSA-FITC, no fluorescence was seen under fluorescence microscopy. When treated with 25 μg/ml P-BSA-FITC, cells showed significant time-dependent elevation in labeled cells at the initial 60 minutes, and then the labeled cells remained at a high level,with no difference between 60 minutes and 120 minutes. Conclusion There are progesterone receptors on the membrane of endometrial carcinoma cell line Ishikawa. The ability of membrane receptor to bind to progesterone is time and dose related.
