摘  要:目的探讨不同浓度脂多糖(LPS)对JEG-3细胞系凋亡的影响,以阐明LPS在妊娠期高血压疾病发病中的作用。方法采用流式细胞术检测JEG-3细胞凋亡率,荧光染色和透射电镜观察滋养细胞凋亡的数目变化、细胞形态和超微结构。结果荧光染色及电镜观察到LPS处理后滋养细胞染色体浓缩,细胞核裂解,见凋亡小体等典型凋亡表现。25、50、100ng/ml的LPS处理后JEG-3细胞的凋亡率分别为(2.06±0.33)%、(7.44±0.68)%、(13.44±0.90)%,显著高于对照组(P均<0.01)。结论LPS能够诱导滋养细胞过度凋亡,且凋亡率随着LPS浓度的增加而增加。Objective To investigate the effect of hpopolysaccharide(LPS) on apoptosis of trophoblasts and their role in the pathogenesis of hypertensive disorder complicating pregnancy. Methods With the treatment of LPS with different concentrations and observed the morphologic change and uhrastnlctural of JEG-3 trophoblasts by fluorescence microscopy and transmission electron microscopy (TEM) and the apoptosis rate of trophoblasts were determined by flow cytometry (FCM). Results observed typical changes of apoptosis (chromosome condensation, karyorrhexis,apoptotic body ) by TEM and Fluorescence microscopy. The munber of apoptosis was increased with elevation of LPS concentration under the microscope, the apoptosis rate of trophoblasts by flow cytometry (FCM) were 2.06+0.33% ,7.44+0.68%, 13.44+ 0.90%, which were significantly higher than the control group (P 〈 0. 01 ). Conclusion LPS can induce excess apoptosis of trophoblasts with elevation of LPS concentration and it is the theory of guiding clinical treatment of hypertensive disorder complicating pregnancy.
