摘 要:目的探讨胸苷酸合成酶(thymidylate synthase,TS)mRNA和二氢嘧啶脱氢酶(dihydropyrimidine dehydrogenase,DPD)mRNA在癌组织和外周血中的表达及其相互关系。方法采用RT-PCR检测36例消化道肿瘤及外周血中TSmR-NA和DPDmRNA的表达水平,同时检测32例对照组的外周血表达水平。各指标之间相关因素的差异性比较采用χ2检验,相关性研究采用Bivariate相关分析。结果在癌组织中TSmRNA检出52.8%(19/36),DPDmRNA检出44.4%(16/36),高于外周血的检出率30.5%和25.0%,但差异无统计学意义(P>0.05)。肿瘤组织TSmRNA、DPDmRNA和外周血的表达结果呈正相关,r值分别为0.627和0.645,在外周血中肿瘤组的表达高于对照组(P<0.01)。肿瘤组织中TSmRNA、DPDmRNA表达水平与细胞分化类型无关,P>0.05。而与TNM分期有关,Ⅲ-Ⅳ期表达水平高于Ⅰ-Ⅱ期表达水平,P<0.05。结论外周血和肿瘤组织中的TSmRNA和DPDmRNA呈正相关,用RT-PCR方法检测外周血TSm-RNA和DPDmRNA,有助于临床对肿瘤的分期判断和用药的选择。Objective To investigate the expression levels of thymidylate synthase (TS)mRNA and dihydropyrimidine dehydrogenase (DPD)mRNA in tumor tissues and the peripheral blood and their relationships.Methods RT-PCR was used to determine the expression levels of TS-mRNA and DPD-mRNA in gastroimestinal tumor tissues and the peripheral blood of 36 patients simultaneously the levels of two enzymes in the peripheral blood of 32 health adults were tested. The data were processed by chi-square test and the correlation was analyzed by Bivariate. Results The expression levels of TS-mRNA and DPD-mRNA in gastrointestinal tumor tissues (52.8 % and 44.4 % ) ) were higher than those in the peripheral blood (30,5 % and 25.0 % ), but there was no statistical significant of them(P 〉 0,05). The levels of TS-mRNA and DPD-mRNA in tmnor tissue had positive correlation with those in the peripheral blood. The R value were 0.627 and 0. 645 respectively. In the peripheral blood, the expression levels of TS-mRNA and DPD-mRNA in tumor groups were higher than those of healthy controls ( P 〈 0.01) .The levels of TS-mlLNA and DPD - mRNA in tumor tissue had no relation with the type of the cell differentiation ( P 〉 0.05) ,but they had relation with the TNM stages. The levels in TNM stageⅢ -Ⅳtumor were higher than those in the fissue of TNM stage Ⅰ - Ⅱtumor(P〈0.05).Condusion The levels of TS mRNA and DPDrnRNA in the tumor tissues had positive correlation with those in the peripheral blood. The detection of TS mRNA and DPDmRNA in the peripheral blood by RT-PCR is helpful to diagnose and judge the classification of the tumor as well as the medicine choice.
