摘  要:目的检测精神分裂症患者免疫型一氧化氮合酶(iNOS)和内皮型一氧化氮合酶(eNOS)的基因表达水平,探讨其与临床症状之间的关系。方法采用逆转录-聚核酶链反应(RT—PCR)和半定量技术,分别检测63例精神分裂症患者,54例同胞对照和51例正常对照外周血单个核细胞iNOS、eNOS的基因表达水平,同时应用阳性与阴性症状量表(PANSS)评定精神分裂症患者的临床症状。结果精神分裂症患者组、同胞对照组和正常对照组的iNOS基因表达水平分别为(1.61±0.10),0.00和0.00,eNOS基因表达水平分别为(1.61±0.11),(1.59±0.11)和(0.60±0.17)。其中iNOS基因在对照组(包括同胞对照和正常对照)未见表达;患者组和同胞对照组的eNOS基因表达水平均显著高于正常对照组(P〈0.05)。同时发现iNOS的基因表达水平与PANSS量表的阳性症状总分显著相关(r=0.462,P〈0.01)。结论1.精神分裂症患者存在iNOS的过度表达,而iNOS可能通过产生大量的NO,参与精神分裂症阳性症状的形成。2.遗传背景在精神分裂症的eNOS基因表达方面可能发挥着重要作用。Objective To investigate the gene expression levels of nitric oxide synthase of schizophrenic, and explore the relation of gene expression levels and the score of Positive and Negative Syndrome Scale( PANSS). Methods The clinical symptom of 63 schizophrenic patients were evaluated by PANSS, and the gene expression levels of inducible nitric oxide synthase ( iNOS ), endothelial nitric oxide synthase ( eNOS ) in peripheral blood mononuclear cell were measured with RT-PCR and semi-quantification technique. 54 sibling controls and 51 normal controls also received gene expression measurements. Results The gene expression levels of iNOS was 1.61 ± 0.10 in schizophrenic ,0.00 in sibling controls and normal controls;the gene expression levels of eNOS was 1.61 ±0.11 in schizophrenic,1.59 ±0. ll in sibling controls and 0.60 ±0.17 in normal controls. No gene expression of iNOS was found in sibling controls and normal controls. The gene expression levels of eNOS was higher in schizophrenic or sibling controls than normal controls (P 〈 0.05 ). The correlation between the scores of positive syndrome in PANSS and the gene expression levels of iNOS was significant respectively (r = 0. 462, P 〈 0.01 ). Conclusion iNOS have over expressions in the peripheral blood mononuclear cell of schizophrenic and may have relationship with the positive syndrome in schizophrenic. Hereditary factor maybe plays a very important role in the regula- tion of the gene expression evels of eNOS.
