摘  要:目的观察烹调油烟冷凝物(COFC)诱发细胞产生活性氧(ROS)及二甲基亚砜(DMSO)的保护作用,为烹调油烟致癌机制以及二甲基亚砜可能的防癌作用提供参考依据和线索。方法以人支气管上皮细胞(BEAS-2B)为靶细胞,细胞上清中过氧化氢(H2O2)、超氧阴离子(O2.-)和羟自由基(.OH)含量分别用辣根过氧化物酶介导的酚红氧化法、细胞色素C还原法和番红花红褪色法进行测定;细胞内H2O2和O2.-分别用2,7-二氯荧光黄双乙酸盐(DCFH2-DA)和氢化乙锭(HE)标记,荧光法测定;自动生化仪测量乳酸脱氢酶。结果BEAS-2B细胞经COFC染毒后,细胞内和细胞上清中ROS含量显著增高,引起细胞内的乳酸脱氢酶溢出;0.5%的DMSO对烹调油烟冷凝物诱发BEAS-2B细胞产生的ROS有明显的抑制作用。结论烹调油烟冷凝物能造成细胞的氧化损伤,可能是烹调油烟致癌机制之一;DMSO对烹调油烟冷凝物所致细胞的氧化损伤具有保护作用,提示DMSO可能通过抑制烹调油烟诱导的氧化损伤,抑制和对抗可能由氧化损伤机制导致的肺癌。Objective To observe the production of reactive oxygen species (ROS) in human bronchial epithelial cells (BEAS-2B) induced by cooking oil fume condensation (COFC) and the protection by dimethyl sulphoxide (DMSO) against the oxidative damage. Methods Extracellular superoxide anions (O2· - ) was measured by the ferricytochrome c reduction method, quantitation of extracellular hydrogen peroxides (H2O2) by the horseradish peroxidase-dependent oxidation of phenol red method, and extracellular hydroxyl radicals (·OH) by decoloration of safranine T. Both O2· and H2O2 were determined by the fluorometric methods, and LDH was detected by auto- biochemistry analyzer. Results The ROS production including H2O2, O2· - and ·OH induced by COFC in BEAS- 2B cells increased significantly, and these increases could be effectively inhibited by 0.5 % of DMSO. Conclusions COFC causes oxidative damage to BEAS - 2B cell, which suggests that the oxidative damage is likely to be one of mechanisms of lung cancer caused by COFC. The oxidative damage to BEAS - 2B cell can be protected by DMSO, which cues that DMSO could inhibit and antagonize the cancer caused by COFC through the oxidative damage mechanism by any possibility.
