摘  要:目的:探讨三磷酸肌醇(IP3)和Fas基因表达变化在genistein诱导肝癌细胞凋亡中的作用。方法:以肝癌HepG2细胞培养72h为对照,以20、40、60、80μmol/L genistein作用于HepG2细胞72h和60μmol/L genistein作用于HepG2细胞6、12、24、48、72h,应用同位素试剂盒IP3-[3H] Birtrak检测细胞IP3含量,RT-PCR分析Fas mRNA表达,Western blotting分析细胞Fas蛋白表达,流式细胞仪检测细胞凋亡率。结果:各浓度的genistein作用于肝癌HepG2细胞72h,IP3含量显著低于对照组,Fas mRNA表达显著高于对照组,Fas蛋白表达显著高于对照组,细胞凋亡率显著高于对照组。60μmol/L genistein作用于肝癌HepG2细胞6、12、24、48、72h,6h后IP3含量显著低于对照组;12h后Fas mRNA表达显著高于对照组,6h后Fas蛋白表达显著高于对照组,24h后各时相细胞凋亡率显著高于对照组(均P<0.01)。结论:Genistein能减少IP3生成,上调Fas基因表达,诱导肝癌细胞凋亡。Objective To investigate the roles of 1, 4, 5- trisphosphate inositol (IP3) and Fas gene expression in apoptosis of HepG2 cells induced by genistein. Methods HepG2 cells Were incubated with 20,40,60,80 μmol/L genistein for 72 h, and incubated with 60 μmol/L genistein for 6, 12, 24, 48 and 72 h, respectively, The level of IP3, Fas expression and apoptosis rate were determined by IP3-[^3 H ] Birtrak assay, RT-PCR, Western blotting and flow cytometry, respectively. Results To the HepG2 cells incubated with each concentration of genistein for 72 h, the IP3 level was lower than that of control group, the Fas mRNA expression was higher than that of control group, the Fas protein expression was higher than that of control group, the apoptosis rate was higher than that of control group. To the HepG2 cellls incubated with 60 I^mol/L genistein for 6, 12, 24, 48 and 72 h, the IP3 concentration was lower than that of control group, the Fas mRNA expression was higher than that of control group incubated with 60 μmol/L genistein for over 12 h, the Fas protein expression was higher than that of control group incubated with 60 μmol/L genistein for over 6 h, the apoptosis rate was significantly higher than that of control group incubated with 60 μmol/L genistein for 24, 48 and 72 h(all P 〈 0.01). Conclusion Genistein can induce apoptosis of HepG2 cells by reducing IPs production and Fas gene expression.
