摘 要:目的:通过两种免疫策略比较三类细菌基因组和CpG ODN对禽流感病毒(AIV)H5N1灭活油乳苗的影响。方法:苯酚氯仿法抽提革兰氏阴性致病菌(大肠杆菌O2)、阳性致病菌(金黄色葡萄球菌)和阳性益生菌(粪链球菌FQ68)的基因组,并人工合成CpG ODN作为AIV油乳苗的佐剂,分佐剂与油苗分开和油裹于油苗内两种途径免疫鸡群,检测血凝(HI)抗体效价、HA抗原特异性IgG效价、IL-10和IFN-γ浓度。结果:当佐剂与油苗分开免疫时,各佐剂在所有的时间点上削弱(P>0.05)或者显著地削弱(P<0.05)HI和血清IgG效价,稍稍提高IL-10和IFN-γ浓度(P>0.05);当佐剂油裹于油苗里时,CpG ODN和FQ68基因组在所有的时间点上提高(P>0.05)或者显著地提高(P<0.05)HI和IgG效价以及IFN-γ浓度,其中CpG ODN好于FQ68基因组,但是都降低了IL-10浓度。结论:当CpG ODN或FQ68基因组油裹在AIV油乳苗里时能提高体液、Th1型免疫水平;当佐剂和油苗分开免疫时,则免疫佐剂性不佳。Objective:To investigate the immunoadjuvant effect induced by three types of bacterial genomes and CpG ODN to inactivated oil emulsion vaccine against Avian Influenza Virus(AIV) H5N1 with two different immune strategies.Methods:The genomes of Gram-negative pathogenic bacterium(E.coli,O2),Gram-positive pathogenic bacterium(Staphylococcus aureus) and Gram-positive probiotics(Streptococcus faecalis,FQ68) were extracted with the phenol-chloroform method.The genomic DNA or synthetic CpG ODN were used as adjuvants in AIV oil emulsion vaccine.There were two ways designed for immunization:the adjuvants were separated from or packed into oil emulsion vaccine;the hemagglutination inhibition(HI) titre,H5 antigen-specific IgG,and the concentrations of interferon-γ(IFN-γ) and IL-10 were examined.Results:When adjuvants were separated from oil emulsion vaccine,each adjuvant reduced(P〉0.05) or significantly reduced(P〈0.05) HI titres and sera IgG titres,slightly raised the concentrations of IFN-γ and IL-10(P〉0.05) at all time points.When adjuvants were packed into oil emulsion vaccine,CpG ODN and FQ68 genome enhanced(P〉0.05) or significantly enhanced(P〈0.05) HI titres,IgG titres and IFN-γ concentration,with decreased IL-10 concentration(P〉0.05).In this case,the immunoadjuvant capability of CpG ODN was better than FQ68 genomes.Conclusion:CpG ODN or FQ68 genome enhances humoral and Th1 type of immune responses when packed into AIV oil emulsion vaccine,while the adjuvants do not show better adjuvanticity when separated from the oil emulsion vaccine.
