摘  要:目的探讨低频超声对大鼠C6胶质瘤细胞内肿瘤坏死因子-α(TNF-α)表达水平的影响,为进一步研究低频超声开放血脑肿瘤屏障的机制提供实验依据。方法应用1MHz低频超声辐照体外培养C6胶质瘤细胞后,采用酶联免疫吸附试验(ELISA)和逆转录-聚合酶链反应(RT-PCR)法分别检测细胞培养液中TNF-α的含量及胶质瘤细胞中TNF-α mRNA的表达。结果ELISA结果显示C6胶质瘤细胞在低频超声辐照后1.5h~3h,其细胞培养液中TNF-α的含量达高峰。RT-PCR结果亦证明了低频超声辐照后1.5h~3h,TNF-α mRNA的表达水平与对照组相比达高峰。结论低频超声辐照可刺激大鼠C6胶质瘤细胞TNF-α分泌增加,TNF-α的表达上调可能在低频超声开放血肿瘤屏障的过程中发挥一定作用。Aim To investigate the effects of low frequency ultrasound treatment on the expression of tumor necrosis factor-α(TNF-α) in C6 glioma cells and provide an experimental basis for the mechanism of opening the blood-brain tumor barrier. Methods ELISA and RT-PCR were used to detect the content of TNF-α in the culture media and the expression of TNF-α mRNA in C6 glioma ceils was treated with 1 MHz low frequency ultrasound respectively. Results ELISA revealed that the content of TNF-α in the cell culture media reached peak value with low frequency ultrasound treated for 1.5 - 3 hour. RT-PCR also revealed that the expression of TNF-α mRNA was the most significantly elevated campared to that of the control group with low frequency ultrasound treated for 1.5 - 3 hour. Conclusion The secretion of TNF-α could be increased in C6 glioma cells induced low frequency uhrasound treatment, which might play an important role in the process of low frequency ultrasound treatment opening the blood-brain tumor barrier.
